淋巴结组织术中快速冷冻切片的优化 被引量:4

摘要 目的优化淋巴结组织术中快速冷冻切片的条件,以期提高淋巴结组织冷冻切片的时效性,改善冷冻切片的质量。方法收集厦门大学附属第一医院病理科2018年1月至2019年7月接收的120例术中淋巴结组织标本。根据组织取材大小不同分为3组,每组随机抽取一定数量的冷冻头,从组织冷冻前是否运用"吸水吸油法"、小淋巴结组织的摆放方式及冷冻切片机样本头的温度设置3个方面分别进行对比实验,比较不同条件对淋巴结组织的冷冻制片时间及切片质量(无皱褶、无裂隙、无冰晶3个指标)的影响。结果冷冻前先将淋巴结组织进行"吸水吸油"操作、将多枚小淋巴结组织并列排列、设置冷冻切片机样本头温度为-20℃的条件下淋巴结组织冷冻切片制片时间最短,无皱褶、无裂隙、无冰晶3个指标得分最高,切片质量最佳。结论优化淋巴结组织"吸水吸油"操作、小淋巴结组织的摆放方式、冷冻切片机样本头的温度设置等条件,可以显著缩短淋巴结组织冷冻制片时间,提高切片质量,值得推荐。 Objective To optimize the conditions of intraoperative frozen sections of lymph node tissue,so as to improve the timeliness and quality of frozen sections of lymph node tissue.Methods 120 intraoperative lymph node tissue specimens were collected from the department of pathology,the first affiliated hospital of xiamen university from January 2018 to July 2019.The specimens were divided into three groups according their different sizes,and a certain number of specimens on frozen heads were randomly selected from each group.Comparative experiments were performed from three aspects:whether to use the"water absorption and oil absorption method"before tissues being frozen,the arrangement of small lymph node tissues and the temperature setting of the sample freezing head of cryostat microtome,so as to compare the effects of different conditions on the frozen section preparation time of lymph node tissues and the slice quality(based on three indicators:no wrinkles,no cracks,no ice crystals).Results The conditions for the shortest preparation time and the best section quality of lymph node tissue(with the highest score of the three indexes:no wrinkles,no cracks and no ice crystals)were as follows:the lymph node tissue should be operated with"water absorption and oil absorption"before being frozen,several small lymph node tissues should be arranged in parallel,and the temperature of the sample head of freezing microtome should be-20℃.Conclusion Optimizing the operation of"water absorption and oil absorption"of lymph node tissues,the way of placing small lymph node tissues,and the temperature setting of sample head of the freezing microtome,can significantly shorten the frozen section preparation time and improve the quality of slicing of lymph node tissues,which is worth recommending.
