摘要
目的:探讨桑白皮各性味拆分组分的寒热药性。方法:雄性SD大鼠连续灌胃优甲乐(120μg/kg)15 d建立热证模型,同时给药桑白皮水煎液及各性味拆分组分,给药结束后记录各组大鼠体质量、肛温及自主活动次数,采用ELISA法检测肝脏中GCK、PFK-1、PK、PGK、PDH、acetyl-CoA、ICD、α-KGDHC、PYGL、GSK-3、CCR、COX、ATPs、ADK、Na+-K+-ATP酶的表达及ATP、ADP、AMP、NAD+、NADH的含量。结果:桑白皮水煎液及各性味拆分组分均升高体质量,降低肛温,减少自主活动次数,降低GCK、PFK-1、PK、PGK、PDH、acetyl-CoA、CS、ICD、α-KGDHC、FUM、PYGL、GSK-3、ATGL、ATP、CCR、COX、ATPs、ADK、Na+-K+-ATP、NADH的表达或含量,升高ADP、NAD+、NAD+/NADH的含量。结论:桑白皮各性味拆分组分能显著改善热证的亢盛状态,其药性为“寒凉。”
Objective:To explore the cold and hot medicinal properties of the various components of Mori Cortex.Methods:Male SD rats were fed with 120μg/kg for 15 consecutive days to replicate the heat syndrome model.The establishment of the model and administration of Mori Cortex and its split componentsat the same time,recorded during the take medicine of the weight of the rats,body temperature and locomotor activity to the state after the end of take medicine,and ELISA method was used to detect the levels of GCK,PFK-1,PK,PGK,PDH,acetyl-CoA,ICD,α-KGDHC,PYGL,GSK-3,CCR,COX,ATPs,ADK,Na+-K+ATP,ATP,ADP,AMP,NAD+,NADH in the liver.Results:The decoction of Mori Cortex and the components of its flavor can increase body weight,lower the anal temperature and decrease the number of voluntary activities,and could reduce the expressions of GCK,PFK-1,PK,PGK,PDH,acetyl-CoA,CS,ICD,α-KGDHC,FUM,PYGL,GSK-3,ATGL,ATP,CCR,COX,ATPs,ADK,Na+-K+ATP,NADH,increase the contents of ADP,NAD+,NAD+/NADH.Conclusion:The properties of Mori Cortex can significantly improve the state of hyperactivity of heat syndrome model,and further confirm that the components of Mori Cortex are the"Cold and cool".
