摘要
目的探讨过表达FOSL1基因对骨髓间充质干细胞(MSCs)增殖与迁移的影响。方法实验分为转基因组(实验组)和MSCs组(对照组)。将FOSL1基因转染MSCs,通过Real-time PCR与Western Blot验证转染效果;通过CCK8增殖实验、PI染色实验和划痕实验探讨MSCs增殖与迁移功能的改善。结果 Real-time PCR与Western Blot实验显示实验组FOSL1在基因与蛋白水平高于对照组(P<0.05)。CCK8增殖实验结果发现实验组增殖率高于对照组(P<0.05)。PI染色实验结果发现实验组G0/G1期细胞占比低于对照组,而S期与G2/M期的细胞比例高于对照组(P<0.05)。划痕实验结果显示实验组的迁移距离大于对照组(P<0.05)。结论 FOSL1基因转染可改善MSCs体外增殖与迁移能力。
Objective To explore the effect of FOSL1 gene overexpression on the proliferation and migration of bone marrow mesenchymal stem cells(MSCs). Methods 2 groups were set up in the study: experiment group(transgene group)and control group(MSCs group);MSCs were transfected with FOSL1 gene, and the transfection efficiency was verified by Real-time PCR and Western Blot assay. CCK8 proliferation assay, propidium iodide(PI) staining and scratch adhesion test were applied in detecting the improvement of proliferation and migration of MSCs after transfection. Results Real-time PCR and Western Blot assays showed that the m RNA and protein expression level of FOSL1 gene in experimental group were obviously higher than those in control group(P<0.05). CCK 8 proliferation assays revealed that the proliferation rate in experiment group was significantly higher than that in control group(P<0.05). PI staining revealed that the ratio of G0/G1 phase cells in experiment group was obviously lower than that in control group while the ratios of the S and G2/M phase cells in experimental group were obviously higher than that in control group(P<0.05). Scratch adhesion test showed that the migration distance in experimental group was longer than that in control group(P<0.05). Conclusions FOSL1 gene transfection may improve the proliferation and migration of MSCs in vitro.
