摘要 糖原合成酶激酶-3β(glycogen synthase kinase-3β,GSK-3β)是一种广泛表达的活性丝氨酸/苏氨酸激酶,通过多种信号途径使细胞底物磷酸化,从而调节多种细胞功能,包括发育、代谢、基因转录、蛋白翻译、细胞骨架组织、细胞周期调控和凋亡等。大量研究发现,GSK-3β在心脏中发挥着重要的调控作用,如调节心脏发育、心肌细胞增殖、心肌肥厚、心肌纤维化等一系列生理病理过程。值得注意的是,心肌纤维化作为心肌损伤的代偿性反应,几乎每一种类型的心脏疾病都与心肌纤维化有关,而GSK-3β被认为是心肌纤维化的重要调节因子。因此,本文主要对GSK-3β调节的心肌纤维化在不同信号转导通路和心脏各种疾病中的作用进行综述。 Glycogen synthase kinase-3β(GSK-3β)is a widely expressed active serine/threonine kinase,which phosphorylates cell substrates through a variety of signal pathways to regulate a variety of cellular functions,including development,metabolism,gene transcription,protein translation,cytoskeleton tissue,cell cycle regulation,and apoptosis.A large number of studies have found that GSK-3βplays an important regulatory role in heart by participated in a series of physiological and pathological processes,such as cardiac development,myocardial proliferation,myocardial hypertrophy,myocardial fibrosis and so on.It is noticeable that,as a compensatory response to myocardial injury,myocardial fibrosis is related to almost every type of heart disease,and GSK-3βis considered to be an important regulator of myocardial fibrosis.Therefore,this paper mainly reviews the role of myocardial fibrosis regulated by GSK-3βin different signal transduction pathways and different cardiac diseases.
