神经生长因子对鼻-鼻窦炎模型大鼠的治疗作用及机制研究

摘要 目的探讨神经生长因子(nerve growth factor,NGF)对鼻-鼻窦炎模型大鼠的作用和机制。方法30只SD大鼠随机分为正常组、模型组及NGF组,每组10只,模型组及NGF组建立急性鼻-鼻窦炎模型,NGF组每天经鼻给予NGF治疗3周。取鼻腔灌洗液计算炎性细胞数量和干扰素γ(IFN-γ)、白细胞介素4(IL-4)含量;取鼻窦黏膜组织HE染色检测病理变化,免疫组化检测IFN-γ、IL-4表达分布;Western blot及RT-PCR检测组织水平IFN-γ、IL-4表达变化。结果NGF组鼻腔灌洗液中嗜酸细胞百分比(Eos%)(7.67±1.49)%显著低于模型组(P<0.05);NGF组鼻黏膜病理变化较模型组显著改善,NGF组IFN-γ含量(202.69±14.35)pg/ml较模型组增加(P<0.05),IL-4蛋白含量(143.00±13.19)pg/ml较模型组显著降低(P<0.05);3组大鼠鼻窦组织中IFN-γ、IL-4 mRNA含量无差异(P>0.05)。结论NGF可能通过促进IFN-γ表达、抑制IL-4分泌抑制Eos活化,发挥对鼻-鼻窦炎模型大鼠的治疗作用。 OBJECTIVE To investigate the effect and mechanism of nerve growth factor(NGF)on rhinosinusitis model rats.METHODS Thirty SD rats were randomly divided into normal group,model group and NGF group with 10 rats in each group.The acute rhinosinusitis model was established in the model group and NGF group.The NGF group was treated with NGF through the nose for 3 weeks every day.Nasal lavage fluid was used to calculate the number of inflammatory cells and the content of IFN-γand IL-4;HE staining was used to detect pathological changes,immunohistochemistry was used to detect the expression of IFN-γand IL-4;Western blot and RT-PCR were used to detect the expression of IFN-γand IL-4 at tissue level.RESULTS The percentage of Eos in nasal lavage fluid of NGF group was significantly lower than that of model group(7.67±1.49)%(P<0.05).The pathological changes of nasal mucosa in NGF group were significantly improved compared with model group.The content of IFN-γin NGF group(202.69±14.35)pg/ml was higher than that in model group(P<0.05),and the content of IL-4 protein(143.00±13.19)pg/ml was significantly lower than that in model group(P<0.05).There was no difference in the content of RNA(P>0.05).CONCLUSION NGF may play a therapeutic role in rats with rhinosinusitis by promoting the expression of IFN-γ,inhibiting the secretion of IL-4 and inhibiting the activation of Eos.
