摘要
目的分析肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)和β1,4-半乳糖基转移酶-Ⅰ(β1,4-Galactosyltransferase-Ⅰ,β1,4-GalT-Ⅰ)在膝关节滑膜炎中的表达及临床意义。方法将2016年11月—2019年11月收治的109例膝关节滑膜炎作为研究组,选取同期进行膝关节探查结果正常者57例作为对照组。观察2组滑膜组织中β1,4-GalT-Ⅰ及TNF-α的表达情况,分析不同剂量、不同时长TNF-α刺激对滑膜组织成纤维样滑膜细胞中β1,4-GalT-Ⅰ表达的影响。结果研究组TNF-α、β1,4-GalT-Ⅰ表达水平高于对照组(P<0.05)。研究组β1,4-GalT-Ⅰ的表达水平随TNF-α剂量、刺激时长的增加不断上升,在5 ng/ml TNF-α时达到顶峰,TNF-α刺激4 h时达到高峰,与对照组比较差异均有统计学意义(P<0.05)。结论膝关节滑膜炎患者滑膜组织中β1,4-GalT-Ⅰ及TNF-α的表达显著增高。不同剂量及不同时长的TNF-α刺激成纤维样滑膜细胞β1,4-GalT-Ⅰ的表达存在差异。
Objective To analyze the expression and clinical significance of tumor necrosis factor-α(TNF-α)and β1,4-Galactosyltransferase-Ⅰ(β1,4-GalT-Ⅰ)in synovitis of knee joint.Methods A total of 109 patients with synovitis of knee joint who were treated in our hospital from November 2016 to November 2019 were enrolled as research group,and 57 healthy people who had normal results of knee joint examination during the same period were selected as the control group.The expressions of β1,4-GalT-Ⅰand TNF-αin synovium of the two groups were observed.The effects of different doses and different durations of TNF-αon the expressions ofβ1,4-GalT-Ⅰin fibroblast-like synovial cells of the synovial tissues were analyzed.Results The expression levels of TNF-αandβ1,4-GalT-Ⅰin the research group were significantly higher than those in the control group(P<0.05).The expressions ofβ1,4-GalT-Ⅰin the research group continued to increase with the increase of the dose and the duration of stimulation,reaching a peak at 5 ng/ml TNF-α.After being stimulated with TNF-α,it reached a peak at 4 h.Compared with the control group,there was a significant difference(P<0.05).Conclusion The expressions of β1,4-GalT-Ⅰand TNF-α in synovial tissue of patients with synovitis of knee joint are significantly higher than those in healthy people.There were significant differences in the expression of β1,4-GalT-Ⅰin fibroblast-like synoviocytes stimulated by different doses and different durations of TNF-α.
