摘要 目的探讨远志药效成分3,6'-二芥子酰基蔗糖(3,6-disinapoylsucrose,DISS)对脑卒中后抑郁(PSD)动物模型的影响.方法建立卒中后抑郁大鼠模型,将30只SD模型造模中的大鼠随机分为PSD+生理盐水组(PSD+NS组)、PSD+氟西汀组(PSD+FLX组)、PSD+3,6'-二芥子酰基蔗糖组(PSD+DISS组),每组各10只,另取10只正常雄性SD大鼠为正常组(CC组),在治疗后行糖水偏好程度测试.第22天以ELISA法测定大鼠前额5-羟色胺(5-HT)、多巴胺(DA)、去甲肾上腺素(NE)浓度.结果在5-HT和DA水平测定中,与CC组比较,除FLX组外,其余PSD组均较低(P<0.05);与NS组比较,DISS、FLX组5-HT和DA水平较高(P<0.05).在NE水平上,FLX组高于CC组(P<0.05),PSD组均低于CC组(P<0.05),且NS组最低;与NS组比较,其余两个PSD组均较高(P<0.05).结论DISS对PSD大鼠有改善作用,并通过作用于单胺递质水平发挥作用. Objective To investigate the effect of active components from Polygala tenuifolia 3,6-disinapoyl sucrose(DISS)on the animal model of post-stroke depression(PSD).Methods 30 SD rats were randomly divided into PSD+saline group(PSD+NS group),PSD+-fluoxetine group(PSD+FLX group),PSD+3,6-disinapoyl sucrose group(PSD+DISS group).Ten normal male SI)rats were divided into normal group(CC group).Sucrose preference was tested after treatment.The concentration of 5-HT,DA and NE in rat forehead was determined by ELISA on the 22nd day.Results The DISS were observed improved the behavioral results of PSD rats.In the determination of monoamine transmitter levels,except the FLX group,the levels of 5-HT and DA in other PSD groups were lower than those in CC group(P<0.05);compared with NS group,the levels of 5-HT and DA in DISS and FLX groups were higher(P<0.05).As for the level of NE,FLX group was higher than CC group and had statistical difference,the other PSD groups were lower than CC group(P<0.05),and NS group was the lowest;compared with NS group,the other two PSD groups were higher(P<0.05).Conclusion DISS can improve the effect of rats with PSD,and plays a role in the level of monoamine transmitters.
