摘要
目的:探讨GATA结合蛋白6(GATA6)和赖氨酰氧化酶样蛋白2(LOXL2)在肾细胞癌(RCC)组织中的表达及其临床意义。方法:选取本院肿瘤科收治的64例肾细胞癌患者作为研究对象,收集手术中的癌组织(研究组)和相匹配的癌旁组织(对照组),采用实时荧光定量PCR(qRT-PCR)方法检测两组患者组织中的GATA6和LOXL2表达水平,观察比较二者表达与临床病理参数的关系,二者表达相关性及对预后的影响。结果:研究组患者组织中GATA6和LOXL2表达水平均高于对照组,两组比较差异有统计学意义(P<0.05);GATA6和LOXL2表达均与年龄、病理分级无关(P>0.05),与病理分期、临床分期、淋巴结转移有关(P<0.05);通过Pearson相关性分析显示,RCC患者GATA6和LOXL2表达呈正相关性(P=0.000);GATA6高表达组患者中位生存时间为43.179(37.369~48.990)个月,GATA6低表达组患者中位生存时间为50.295(45.481~55.110)个月,两组比较差异有统计学意义(P=0.000);LOXL2高表达组患者中位生存时间为41.019(34.319~47.719)个月,LOXL2低表达组患者中位生存时间为48.388(42.759~54.017)个月,两组比较差异有统计学意义(P=0.000)。结论:GATA6和LOXL2在RCC组织中均表达上调,并与病理分期、临床分期、淋巴结转移有关,两者表达呈正相关,GATA6和LOXL2高表达患者中位生存时间短,预后较差。
Objective:To investigate the expressions and clinical significances of GATA binding protein 6(GATA6)and lysine oxidase like protein 2(LOXL2)in renal cell carcinoma(RCC)tissues.Methods:Sixty-four patients with renal cell carcinoma were selected as the subjects.The cancer tissues(study group)and matched adjacent tissues(control group)were collected during operation.The expression levels of GATA6 and LOXL2 in two groups were detected by real-time fluorescence quantitative PCR(qRT-PCR).The relationships between the two expressions and clinical parameters,and the correlation between the two expressions and prognosis were observed and compared.Results:The expression levels of GATA6 and LOXL2 in the study group were higher than those in the control group,and there was significant difference between the two groups(P<0.05).The expressions of GATA6 and LOXL2 were not correlated with age and pathological grade(P>0.05),but with pathological stage,clinical stage and lymph node metastasis(P<0.05).Pearson correlation analysis showed that there was a positive correlation between GATA6 and LOXL2 expression in RCC patients(P=0.000).The median survival time was 43.179(37.369~48.990)months in the high GATA6 expression group and 50.295(45.481~55.110)months in the low GATA6 expression group,and the difference between the two groups was statistically significant(P=0.000).The median survival time was 41.019(34.319~47.719)months in the high LOXL2 expression group,and 48.388(42.759~54.017)months in the low LOXL2 expression group,the difference between the two groups was statistically significant(P=0.000).Conclusion:Both GATA6 and LOXL2 are up-regulated in RCC tissues,are related to pathological stage,clinical stage and lymph node metastasis.The two groups are positively correlated.Patients with high expressions of GATA6 and LOXL2 have short survival time and poor prognosis.
