MMP-9与TRAIL在结直肠癌的表达及对术后化疗的评估意义

Expression of MMP-9 and TRAIL in colorectal cancer and evaluation of postoperative chemotherapy

目的检测MMP-9,TRAIL在结直肠癌中的表达情况,探讨二者在癌细胞浸润、转移中的可能相互关系及对术后化疗预后的影响。方法将2010年12月-2014年6月期间在我院行结直肠癌切除术治疗的60例患者作为研究对象,应用组织芯片技术结合免疫组化SP法检测结直肠癌切除标本中MMP-9,TRAIL表达情况,分析其与临床病理指标的关系及对术后化疗预后的影响。结果MMP-9在结直肠癌组织中的表达明显高于癌旁组织(P<0.01),而TRAIL却低于癌旁组织(P<0.05);MMP-9高表达组和TRAIL低表达组中结直肠癌细胞低分化程度所占比重较高,且Ⅲ/Ⅳ期患者所占比重较大,差异均有统计学意义(P<0.05);MMP-9低表达组术后化疗5年生存率显著高于高表达组(P<0.05),而TRAIL低表达组却低于高表达组(P>0.05)。结论结直肠癌组织中MMP-9与TRAIL可能存在拮抗作用,MMP-9高表达与TRAIL低表达预示术后化疗预后较差。

Objective To detect the expression of MMP-9 and TRAIL in colorectal cancer,and to explore the possible relationship between MMP-9 and TRAIL in cancer cell invasion and metastasis and the effect of postoperative chemotherapy.Methods Sixty patients who underwent colorectal cancer resection in our hospital from December 2010 to June 2014 were enrolled in this study.Tissue microarray technique combined with immunohistochemical SP method was used to detect MMP-9 in colorectal cancer resected specimens.TRAIL expression,analysis of its relationship with clinical pathological indicators and the impact of postoperative chemotherapy prognosis.Results The expression of MMP-9 in colorectal cancer tissues was significantly higher than that in adjacent tissues(P<0.01),while TRAIL was lower than that in adjacent tissues(P<0.05).MMP-9 high expression group and TRAIL low expression group The proportion of colorectal cancer cells with low differentiation was higher,and the proportion of patients with stage III/IV was larger,the difference was statistically significant(P<0.05).The 5-year survival rate of postoperative chemotherapy in MMP-9 low expression group Significantly higher than the high expression group(P<0.05),while the low expression level of TRAIL was lower than the high expression group(P>0.05).Conclusion There may be antagonistic effects of MMP-9 and TRAIL in colorectal cancer tissues.High expression of MMP-9 and low expression of TRAIL indicate poor prognosis of postoperative chemotherapy.
