circFAT1在结直肠癌中的表达及其对肠癌细胞迁移和侵袭的影响

The expression of circFAT1 in colorectal carcinomas and its role on tumor cell migration and invasion

目的探讨circFAT1在结直肠癌中的表达及其对肠癌细胞迁移和侵袭能力的影响。方法选取我院收治的结直肠癌患者85例,采用实时荧光定量PCR(Real-Time PCR)和荧光原位杂交(FISH)技术检测circFAT1在肿瘤组织中的表达,分析其与患者临床病理参数和预后的相关性。通过脂质体转染siRNA,敲低circFAT1在肠癌细胞株LoVo中的表达,利用Transwell小室装置检测转染前后肠癌细胞株迁移和侵袭能力的变化。结果circFAT1在结直肠癌组织中表达高于癌旁正常肠黏膜组织(P<0.05)。在结直肠癌中,circFAT1的表达状态与淋巴结转移及患者预后相关(P<0.05)。体外细胞实验显示,下调circFAT1的表达可显著降低肠癌细胞株迁移和侵袭能力(P<0.05)。结论circFAT1在结直肠癌组织中高表达,且与结直肠癌的恶性演进密切相关。

Objective To investigate the expression of circFAT1 in colorectal carcinomas(CRC),and analyze its role on tumor cell migration and invasion. Method The expression levels of circFAT1 in 85 paired CRC tissues and adjacent non-tumor tissues were explored using Real-Time PCR and fluorescent in situ hybridization. The siRNAs targeting circFAT1 were transiently transfected into human CRC cell line LoVo,and the effects of circFAT1 on cell migration and invasion were analyzed by transwell assay. Results Compared with adjacent non-tumor tissues,circFAT1 was significantly up-regulated in CRC tissues(P < 0.05). Increased circFAT1 was significantly associated with lymph node metastasis and poor prognosis(P < 0.05). In vitro,down-regulation of circFAT1 inhibited the migratory and invasive abilities of LoVo cells(P < 0.05). Conclusion circFAT1 might play an important role in the progression of CRC by modulating tumor cell migration and invasion.
