超声波结合聚乙二醇化多西他赛白蛋白纳米粒治疗小鼠乳腺癌模型的研究 被引量:3

摘要 目的研究利用超声波结合聚乙二醇化多西他赛白蛋白纳米粒(PEG-DANPs)治疗小鼠乳腺癌模型,旨在为临床治疗乳腺癌提供一种新思路。方法建立BALB/c小鼠乳腺癌模型,分别用DANPs、PEG-DANPs及PEG-DANPs结合超声波照射对小鼠乳腺癌模型进行治疗,每疗程为1周,共4个疗程,超声照射条件为每次30 s,强度为1 MHz。最后一次给药48 h后,处死全部小鼠,对皮下移植瘤瘤块进行TUNEL法检测癌细胞凋亡情况,取小鼠肺组织及肝组织进行HE染色和病理半定量分析。结果相比单纯PEG-DANPs治疗,超声波结合PEG-DANPs能够更加有效地促进癌细胞凋亡,抑制小鼠乳腺癌的增殖和转移。结论超声波结合PEG-DANPs治疗小鼠乳腺癌模型,疗效明显,可作为治疗乳腺癌的一种新思路。 Objective Breast cancer is the most common cancer among women in China,most patients were diagnosed with advanced stage when discovery and they had to receive chemotherapy only. Therefore,it is urgent to develop a good effect,small toxic effects of chemotherapy drugs. In this study we treated the breast cancer model of mice by ultrasound combined with docetaxel loaded PEG-albumin nanoparticles,aim to provide a new option for clinical treatment of breast cancer. Methods We established the transplantation tumor model of breast cancer. When the tumor volume in transplantation tumor group reached to about 120 mm3,the mice were treated 4 times at 7-day intervals with DANPs,PEG-DANPs and ultrasound combined with PEG-DANPs respectively. The ultrasonic irradiation condition is 30 seconds and the frequency is 1 MHz. 48 hours after the last treatment,the mice were sacrificed. The lung and liver of model were subjected to HE staining and pathological semi-quantitative analysis,the TUNEL method was used to detect apoptosis of cancer cells in transplantation tumor model. Results Compared with simple PEG-DANPs therapy,ultrasound combined with PEG-DANPs can effectively inhibit the growth of breast cancer transplanted tumor,and it can suppress metastases more effectively. Conclusion The antitumor effect is obvious in the treatment of the breast cancer by ultrasound combined with PEG-DANPs,it can be used as a new idea for clinical treatment of breast cancer.
