胎牛血清对猪精子常温负压保存质量的影响

Effect of Fetal Bovine Serum on the Quality of Porcine Sperm under Normal Temperature and Negative Pressure

为探究胎牛血清(FBS)在17℃负压环境下对猪精液的保存效果,延长猪精液保存的时间以及维持猪精子的质量。通过在Modena基础稀释液中分别添加体积分数为0、5%灭活、10%灭活、非灭活5%、非灭活10%的FBS,检测猪精子在此环境下5 d内的精子活率、直线速度、侧摆幅度等为检测参数。结果表明:10%灭活的FBS在猪精子各项参数上都显著优于其他组(P<0.05)。由结果可知,在Modena基础稀释液添加10%灭活的FBS在17℃负压环境下最有利于猪精液的保存,为猪精子的常温保存研究提供实验依据。

To explore the effect of fetal bovine serum(FBS)on the preservation of pig semen under 17℃negative pressure environment,prolong the preservation time of pig semen and maintain the quality of pig sperm,adding the volume fraction in Modena foundation diluent respectively 0,5%inactivated,10%inactivated,5%lived,and 10%lived FBS,detect the pig sperm in the environment within the 5 d’s live sperm rate,linear velocity,side swing range,etc.The results showed that 10%inactivated FBS was significantly better than other groups in various parameters of pig sperm(P<0.05).The result showed that in Modena diluent adding 10%inactivated FBS in 17℃under negative pressure environment most conducive to the pig semen preservation,which would provide experiment basis for the research on the pig sperm normal temperature preservation.
