摘要
目的对virB2基因编码蛋白进行分析,为virB2基因及其编码蛋白功能提供实验依据。方法利用多种生物学软件以及网站对VirB2蛋白的结构和功能进行分析预测,VirB2蛋白序列通过基因推导获得并由生物公司合成,然后通过免疫动物实验制备鼠抗VirB2蛋白多克隆抗体,同时设计进行VirB2蛋白细胞毒试验(MTT法)。结果virB2基因编码蛋白属于疏水性蛋白,为鞭毛样结构,有较强的细胞毒作用。结论对VirB2蛋白的结构和功能进行了分析预测,证明VirB2蛋白在H.pylori相关的致病性特别是引起胃黏膜炎症方面起到一定的作用,能够为研究H.pylori致病机制提供帮助。
Objective To obtain a preliminary understanding of virB2 gene and its coding protein function by biological analysis, prepare polyclonal antibodies and accomplish cytotoxicity test of VirB2 protein. Methods The structure and function of VirB2 protein were analyzed and predicted by using various biological softwares and websites. VirB2 protein sequence was obtained through gene deduction and synthesized by biological companies. Then the mouse anti-VirB2 polyclonal antibody was prepared through immune animal experiments. At the same time, the MTT cytotoxicity test of VirB2 protein was designed and carried out. Results The virB2 gene coding protein was found to belong to hydrophobic protein with flagellate-like structure, which has strong cytotoxic effect. The polyclonal antibodies with high purity were obtained, which laid a foundation for further experiment. Conclusion The analysis of virB2 gene function in the secretion system of H. pylori plasticizable region tfs3 a proves that VirB2 protein plays a certain role in the pathogenicity of H. pylori, especially the gastric mucosal inflammation, and can be used in the study on the pathogenic mechanism of H. pylori.
