川芎嗪对高眼压大鼠氧化应激及视网膜神经节细胞损伤的影响 被引量:8

Effect of ligustrazine on oxidative stress and retinal ganglion cells damage of high intraocular pressure in rats

摘要 目的研究川芎嗪对高眼压大鼠氧化应激及视网膜神经节细胞损伤的影响。方法将30只眼压正常的SD大鼠右眼以烧灼巩膜上静脉法构建高眼压模型,并随机分为模型组与实验组;另选15只眼压正常SD大鼠为对照组。建模1周后实验组大鼠腹腔注射川芎嗪40 mg·kg^-1,模型组与对照组大鼠则腹腔注射等量生理盐水,每天1次,连续干预2周。以笔式眼压计检测大鼠眼压,以黄嘌呤氧化酶法检测超氧化物歧化酶(SOD)含量,以硫代巴比妥比色法检测丙二醛(MDA)含量,以原位末端标记法(TUNEL)检测视网膜组织视网膜神经节细胞凋亡情况。结果干预3周后,对照组、模型组与实验组大鼠眼压分别为(13.61±1.94),(34.97±2.39),(20.02±2.01)mmHg,视网膜组织SOD含量分别为(102.46±9.12),(77.98±8.71),(89.44±7.96)kU·L^-1,MDA含量分别为(2.87±0.89),(8.16±1.21),(4.03±1.08)μmol·L^-1,视网膜神经节细胞凋亡率分别为(13.12±2.13)%,(52.14±3.67)%,(35.08±3.39)%,差异均有统计学意义(均P<0.05)。结论川芎嗪可有效降低高眼压大鼠模型眼压、抑制视网膜组织氧化应激反应及视网膜神经节细胞凋亡。 Objective To explore the effect of ligustrazine on oxidative stress and retinal ganglion cells damage of high intraocular pressure in rats.Methods Thirty cases SD rats with normal intraocular pressure were built high intraocular pressure model by burning episcleral vein method in right eyes,and randomly divided into model group and test group,another 15 SD rats with normal intraocular pressure were selected as control group.At 1 week after modeling,the rats in test group were treated with intraperitoneal injection of 40 mg·kg^-1 ligustrazine,rats in model group and control group were treated with equal amount of normal saline,intraperitoneal injection,one times a day,continuous intervention for two weeks.The intraocular pressure of rats were detected by pen type tonometer,the contents of superoxide dismutase(SOD)of rats were detected by xanthine oxidase method,the contents of malondialdehyde(MDA)of rats were detected by glucosinolates barbitone colorimetry method;the retinal tissue apoptosis of retinal ganglion cells were detected by Terminal deoxynucleotidyl transferase dUTP nick-end labeling(TUNEL).Results The intraocular pressures of rats in control group model group,test group at 3 weeks after intervention were(13.61±1.94),(34.97±2.39),(20.02±2.01)mmH g,the retinal tissue SOD contents were(102.46±9.12),(77.98±8.71),(89.44±7.96)k U·L^-1,MDA contents were(2.87±0.89),(8.16±1.21),(4.03±1.08)μmol·L^-1,retinal ganglion cell apoptosis rate were(13.12±2.13)%,(52.14±3.67)%,(35.08±3.39)%,all with significant difference(all P<0.05).Conclusion ligustrazine can reduce the intraocular pressure of high intraocular pressure model rats,inhibit the retinal tissue oxidative stress and apoptosis of retinal ganglion cells effectively.
