人参皂苷Rg5对多发性骨髓瘤RPMI8226细胞凋亡和周期阻滞的影响及其机制研究 被引量:3

Effect and mechanisms of ginsenoside Rg5 on the apoptosis and cell cycle arrest in multiple myeloma RPMI8226 cells

摘要 目的研究人参皂苷Rg5对多发性骨髓瘤细胞增殖和凋亡的影响及其机制。方法(1)将多发性骨髓瘤细胞分成2组:空白对照组和实验组(110μmol·L^-1人参皂苷Rg5处理),处理培养后2 d;(2)将多发性骨髓瘤细胞分为2组:激活实验组、实验组。激活实验组细胞用110μmol·L^-1人参皂苷Rg5和20 mmol·L^-1 LiCl处理培养,实验组以110μmol·L^-1人参皂苷Rg5培养。以流式细胞术测定细胞周期和凋亡变化,蛋白质印迹法检测细胞中c-Myc蛋白表达水平。结果(1)空白对照组和实验组的细胞凋亡率分别为(5.23±0.41)%和(20.36±1.14)%;这2组的细胞G1期比例分别为(51.84±3.14)%和(67.32±5.11)%;这2组的的细胞β-catenin蛋白水平分别为0.81±0.06和0.39±0.04;这2组的细胞c-Myc蛋白水平分别为0.78±0.07和0.37±0.05。上述指标:实验组和空白对照组比较,差异均有统计学意义(均P<0.05)。(2)实验组和激活实验组的细胞G1期比例分别为(69.48±3.47)%和(56.17±4.27)%;这2组的细胞凋亡率分别为(19.67±1.20)%和(10.35±1.14)%;这2组的细胞c-Myc蛋白水平分别为0.35±0.05和0.70±0.09。上述指标:实验组和空白对照组比较,差异均有统计学意义(均P<0.05)。结论人参皂苷Rg5通过抑制Wnt/β-catenin信号通路,诱导多发性骨髓瘤细胞凋亡和细胞周期阻滞。 Objective To explore the effect and mechanism of ginse-noside Rg5 on proliferation and apoptosis of multiple myeloma cells.Methods(1)Multiple myeloma cells were divided into two groups:blank control group and experimental group(110μmol·L^-1 ginsenoside Rg5 treatment),for 2 days.(2)Multiple myeloma cells were divided into two groups:experimental group and activate experimental group(10μmol·L^-1 Rg5+20 mmol·L^-1 LiCl).Cell cycle and apoptosis were measured by flow cytometry.The expression levels of β-catenin and c-Myc proteins were detected by Western blot.Results(1)The apoptosis rate in blank control group and experimental group were(5.23±0.41)%and(20.36±1.14)%,respectively;the proportion of G1 phase in the two groups were(51.84±3.14)%,(67.32±5.11)%,respectively;the levels of β-catenin protein in the two groups were 0.81±0.06 and 0.39±0.04,respectively;the levels of c-Myc protein in the two groups were 0.78±0.07 and 0.37±0.05 respectively.Comparison between experimental group and blank control group,the difference of the factors were significantly(all P<0.05).(2)The G1 phase ratio experimental group and activate experimental group were(69.48±3.47)%,(56.17±4.27)%,respectively;the apoptosis rate in the two groups were(19.67±1.20)% and(10.35±1.14)%,respectively;the level of c-Myc protein in the two groups were 0.35±0.05 and 0.70±0.09,respectively.The difference of the factors were significantly between experimental group and activate experimental group(all P<0.05).Conclusion Ginsenoside Rg5 inhibits Wnt/β-catenin signaling pathway and induces apoptosis and cell cycle arrest in multiple myeloma cells.
