摘要
目的观察二参颗粒对冠心病心肌缺血大鼠 NF-κB 信号通路中炎症因子的影响。方法 60只健康雄性SD大鼠,随机分为6组:正常组、模型组、二参颗粒高剂量组、中剂量组、低剂量组,冠心生脉丸对照组。于末次灌胃给药前72 h,连续3 d腹腔注射垂体后叶素形成冠心病心肌缺血模型。检测二参颗粒对冠心病大鼠心电图S-T段的影响、采用酶联免疫法测定血清IL-8和IL-6影响、采用实时定量PCR方法分析TLR4、NF-κB含量的影响。结果与模型对照组相比,二参颗粒治疗后二参颗粒高、中、低剂量组各时间点T波的变化值显著降低;与对照组相比,二参颗粒高剂量组、中剂量组、低剂量组均能明显降低大鼠血清IL-8和IL-6含量(P<0.05);冠心病大鼠心脏细胞TLR4和NF-κB基因表达水平明显高于正常组,二参颗粒高、中、低剂量组和冠心生脉丸组对大鼠TLR4和NF-κB基因的表达明显下降,与模型组比较差异显著(P< 0.01)。结论二参颗粒可有效改善冠心病大鼠心律失常,激活NF-κB 信号通路,从而阻止冠心病大鼠炎症反应的发生与发展,充分发挥心肌细胞的营养和再生作用。
Objective To observe the effect of Ershen Granule on inflammatory factors in NF-κB signaling pathway in rats with myocardial ischemia induced by coronary artery disease.Methods A total of 60 healthy male SD rats,randomly divided into 6 groups:normal group,model group,Ershen Granule high,medium and low dose groups,Guanxin Shengmai Pill group.72 hours before the last intragastric administration,the pituitary vasopressin was injected intraperitoneally for 3 consecutive days to form a myocardial ischemia model of coronary artery disease.Effect of Ershen Granule on electrocardiogram S-T segment in rats with coronary artery disease,determination of serum IL-8 and IL-6 by enzyme-linked immunosorbent assay,real-time quantitative PCR was used to analyze the effects of TLR4 and NF-κB levels.Results Compared with the model control group,after the treatment of Ershen Granules,the changes of T wave in the high-dose,medium-dose and low-dose groups of the Ershen Granules were significantly lower;compared with the control group,the high-dose,middle-dose and low-dose groups of Ershen Granules could significantly reduce serum IL-8 and IL-6 levels (P<0.05).The expression levels of TLR4 and NF-κB genes in cardiac cells of coronary artery disease rats were significantly higher than those in normal groups.The expressions of TLR4 and NF-κB genes in the high-dose,medium-dose and low-dose groups of Ershen Granules and Guanxin Shengmai Pill group were significantly decreased,which was significantly different from the model group (P<0.01).Conclusion Ershen Granule can effectively improve arrhythmia in rats with coronary heart disease,activate the NF-κB signaling pathway,thereby preventing the occurrence and development of inflammatory response in rats with coronary heart disease,give full play to the nutrition and regeneration of cardiomyocytes.
