泰山磐石散对复发性流产小鼠T淋巴细胞亚群及妊娠丢失的影响

Effect of Taishan Panshi Powder on T Lymphocyte subsets and pregnancy loss in recurrent abortion mice

目的探讨泰山磐石散结合"预培其损"理论对复发性流产小鼠体内淋巴细胞亚群CD4^+、CD8^+的比例以及妊娠结局的影响。方法采用雌性CBA/J与雄性BALB/c小鼠交配建立正常妊娠小鼠模型,采用雌性CBA/J 小鼠与雄性DBA/2 小鼠交配建立复发性流产小鼠模型,并将流产模型小鼠随机分为4组,分别为复发性流产模型组及泰山磐石散低、中、高剂量组。泰山磐石散低、中、高剂量组从合笼开始灌胃给药,灌胃治疗14 d后处死小鼠,取其脾脏及子宫组织,解剖子宫组织,测量胎盘直径,计算胎盘体积、胚胎丢失率,同时流式细胞仪检测各组小鼠脾脏组织液中淋巴细胞亚群CD4^+、CD8^+的含量并计算2者比例。结果模型组的胚胎丢失率明显高于正常组(P<0.05),与模型组比较,中、高剂量组的胚胎丢失率显著降低(P<0.05);小鼠脾脏组织液中淋巴细胞亚群CD4^+、CD8^+的比例,与正常组相比,模型组小鼠血中CD4^+/CD8^+的比值升高(P<0.05);与模型组比较,泰山磐石散低、中、高剂量组血中CD4^+/CD8^+的比值呈不同程度的降低(P<0.05),且随着剂量的增加,2者比值接近正常组。结论泰山磐石散能使复发性流产小鼠体内淋巴细胞亚群CD4^+/ CD8^+比值向 CD8^+优势转化,诱导妊娠免疫耐受,发挥免疫防护作用,有利于妊娠继续。

Objective To investigate the effects of Taishan Panshi Powder combined with the theory of "prevention and treatment of etiology" on the percentage of CD4^+,CD8^+ lymphocyte subsets in recurrent abortion mice and pregnancy outcomes.Methods Female CBA/J mice were mated with male BALB/c mice to establish normal pregnant mice model.Female CBA/J mice were mated with male DBA/2 mice to establish recurrent spontaneous abortion mice model.The abortion mice were randomly divided into four groups:recurrent spontaneous abortion model group and Taishan Panshi Powder low,medium and high dose group.The mice in low,medium and high dose groups of Taishan Panshi Powder were administered by gavage from cage closure.After 14 days of gavage treatment,the mice were executed.The spleen and uterus tissues were taken,the uterus tissues were dissected,the placenta diameter was measured,the placenta volume and the rate of embryo loss were calculated.At the same time,the content of CD4^+ and CD8^+ lymphocyte subsets in spleen tissue fluid of mice in each group was detected by flow cytometry,and the ratio of CD4^+ and CD8^+ was calculated.Results The rate of embryo loss in the model group was significantly higher than that in the normal group (P<0.05).Compared with the model group,the rate of embryo loss in the middle and high dose groups was significantly lower (P<0.05) after the intervention of traditional Chinese medicine.Compared with the normal group,the ratio of lymphocyte subsets CD4^+,CD8^+ in splenic tissue fluid of mice of the model group was higher (P<0.05).Compared with the model group,the ratio of CD4^+/CD8^+ in blood of the low,middle and high dose groups of Taishan Panshi Powder decreased in varying degrees (P<0.05),and with the increase of dose,the ratio of CD4^+/CD8^+ was close to that of the normal group.Conclusion Taishan Panshi Powder can transform the CD4^+/CD8^+ ratio of lymphocyte subsets in mice with recurrent spontaneous abortion into CD8^+ dominance,induce immune tolerance of pregnancy,exert immune protective effect,which is beneficial to the continuation of pregnancy.
