不同分子量透明质酸对肺癌小鼠体内肿瘤生长转移的影响 被引量:2

摘要 目的观察皮下注射不同分子量透明质酸(HA)对LLC肺癌模型小鼠体内肿瘤生长和转移的影响。方法50只C57BL/6的LLC肺癌小鼠模型随机分成低(HA1)、中(HA2)、高(HA3)分子组,生理盐水组和无注射组,注射上述溶液,每3 d一次,第25 d处死。取肝、肺、脑、肾、肿瘤组织,计算脏体比;HE染色法观察病理学变化;免疫组化法观察胱天蛋白酶3、增殖细胞核抗原(PCNA)、透明质酸合酶2(HAS2)、透明质酸酶2在肿瘤中的表达。结果HA3组远处器官转移率(55.6%)升高,分别向肝、肾、脑转移,尤以肝转移严重(88.9%)。肿瘤组织中,HA3组HAS2阳性表达减弱;HA2组PCNA阳性表达减弱,胱天蛋白酶3阳性表达升高。结论中分子HA抑制肿瘤生长;高分子HA促进肿瘤转移。 Objective To observe the effect of different molecular weight hyaluronic acid(HA)injected subcutaneously on tumor growth and metastasis in LLC lung cancer model mice.Methods Fifty C57BL/6 LLC lung cancer mice were randomly divided into low(HA1),medium(HA2),high(HA3)molecular groups,normal saline group and non injection group.The above solutions were injected once every 3 days,and killed on the 25th day.Liver,lung,brain,kidney and tumor tissues were collected to calculate the ratio of viscera to body;pathological changes were observed by HE staining;expression of caspase-3,proliferating cell nuclear antigen(PCNA),hyaluronic acid synthase 2(HAS2)and hyaluronidase 2 were observed by immunohistochemistry.Results In the HA3 group,the distant organ metastasis rate(55.6%)increased,and the metastasis to the liver,kidney and brain respectively,especially to the liver(88.9%).In tumor tissue,the expression of HAS2 was decreased in HA3 group,PCNA was decreased in HA2 group,and caspase-3 was increased in HA3 group.Conclusion The growth of tumor was inhibited by medium molecular HA,and metastasis was promoted by high molecular HA.
