散结镇痛理气方对原发性肝癌中人解整合素样金属蛋白酶10表达影响的实验研究

Influence of Sanjie Zhentong Liqi Formula on the expression of ADAM10 in primary liver cancer:an experimental study

目的观察散结镇痛理气方对原发性肝癌大鼠体内人解整合素样金属蛋白酶10(ADAM10)表达的影响并探讨其临床效用。方法90只大鼠随机分为正常组、模型组及给药组,每组30只,模型组及正常组以DEN诱导构建大鼠原发性肝癌模型,给药组予散结镇痛理气方汤剂,正常组及模型组则予等量生理盐水灌胃。观测比较肝组织的病理学特征及ADAM10及相关因子在各组大鼠肝脏组织中的表达。结果模型组、给药组ADAM10的阳性表达率显著高于正常组,差异有统计学意义(P<0.05),但给药组在给药干预后则显著降低,差异有统计学意义(P<0.05),且有肿瘤质量体积的缩减,差异有统计学意义(P<0.05),肝细胞变性坏死亦有一定程度改善。结论散结镇痛理气方通过对肝癌组织中ADAM10的表达抑制,阻断肝癌形成与发展进程。

Objective To observe the effect of Sanjie Zhentong Liqi Formula on the expression of A Disintegrin and metalloproteinase domain-containing protein 10(ADAM10)in rats with primary liver cancer and to investigate its clinical efficacy.Methods A total of 90 rats were randomly divided into a normal group,a model group and a treatment group,with 30 rats in each group.DEN-induced rat models with primary liver cancer were established in the model group and the normal group.The treatment group was given the decoction of Sanjie Zhentong Liqi Formula;the normal group and the treatment group were given the same amount of saline.The pathological characteristics of liver tissues and the expression of ADAM10 and related factors of liver tissues in each group were observed and compared.Results The positive expression rate of ADAM10 in the model group and the treatment group was significantly higher than that in normal group(P<0.05),however,the positive rate in the treatment group significantly decreased after intervention(P<0.05),and there was also a reduction in tumor mass and volume(P<0.05),and an improvement in the degeneration and necrosis of liver cells to some extent.Conclusion Sanjie Zhentong Liqi Formula can block the formation and development of liver cancer by inhibiting the expression of ADAM10 in hepatocellular carcinoma tissues.
