摘要
目的研究hsa_circ_0005777在乳腺癌组织中的表达情况,探讨乳腺癌组织中是否存在特异性表达的circRNA及其作为乳腺肿瘤生物学标志物的潜力。方法收集术后乳腺癌患者癌组织和癌旁组织标本23对,提取组织样本RNA,采用qRT-PCR法检测hsa_circ_0005777在乳腺癌及癌旁组织中的表达,ROC曲线分析乳腺癌组织中hsa_circ_0005777表达与患者临床病理因素的关系。并在乳腺癌患者血液样本中验证hsa_circ_0005777的表达。结果与癌旁组织相比,hsa_circ_0005777在乳腺癌组织中下调(P<0.01)。ROC曲线分析结果显示,hsa_circ_0005777在乳腺癌患者组织中的AUC为0.983,灵敏度为95.7%,特异度为95.7%(P<0.01)。hsa_circ_0005777与癌胚抗原具有负相关关系(r=-0.466,P=0.025)。hsa_circ_0005777诊断乳腺癌转移的AUC为0.795,灵敏度为75.0%,特异度为81.8%(P=0.016)。结论hsa_circ_0005777在乳腺癌组织和乳腺癌患者血液中低表达,并与乳腺癌增殖和淋巴结转移以及组织病理分型密切相关,有可能成为乳腺癌治疗靶点和预测乳腺癌进展快慢的一个指标。
Objective To investigate the expression of hsa_circ_0005777 in breast cancer tissues,and to investigate whether circRNAs specifically expressed in breast cancer tissues and their potential as breast cancer biomarkers.Methods A total of 23 pairs of postoperative cancer tissues and paracancerous tissues from breast cancer patients were collected.Total RNA were isolated from the tissue samples.The relative expression of hsa_circ_0005777 in breast cancer and adjacent tissues was detected by quantitative PCR.ROC curve analysis was performed to determine the diagnostic values of candidate hsa_circ_0005777.Expression of hsa_circ_0005777 was verified in blood samples from breast cancer patients.Results Compared with adjacent tissues,hsa_circ_0005777 was significantly down-regulated in breast cancer tissues(P<0.01).ROC curve analysis showed that hsa_circ_0005777 had an AUC of 0.983,a sensitivity of 95.7%,and a specificity of 95.7%(P<0.01).hsa_circ_0005777 was negatively correlated with CEA(r=-0.466,P=0.025).hsa_circ_0005777 diagnosed breast cancer metastasis with an AUC of 0.795,sensitivity of 75.0%,specificity of 81.8%(P=0.016).Conclusion hsa-circ-0005777 is low expression in breast cancer tissue and blood of patients with breast cancer,and is closely related to breast cancer proliferation,lymph node metastasis and histopathological classification,which may become a target of breast cancer treatment and an indicator of breast cancer progress.
