平肝潜阳法方药对高血压病肝阳上亢证大鼠血管组织中AngⅡ和PDGF-BB表达的影响 被引量:12

摘要 目的:观察平肝潜阳法方药对自发性高血压病大鼠(SHR)肝阳上亢证模型血管组织中血管紧张素Ⅱ(AngⅡ)和血小板衍生生长因子-BB(PDGF-BB)表达的影响,探讨平肝潜阳法干预的作用机制。方法:SD大鼠10只为正常组,SHR 20只,灌服附子汤复制成高血压病肝阳上亢证模型,随机分为模型组和治疗组各10只,其中治疗组分别于复制模型成功后给予平肝潜阳法方药干预4周。观察大鼠的行为学和血压变化,酶联免疫法检测AngⅡ和PDGF-BB的含量,采用荧光实时PCR和Western Blot检测各组大鼠血管组织中AT1αR和PDGF-BB基因和蛋白质表达变化。结果:与正常组比较,高血压病肝阳上亢证模型大鼠血压和易激惹程度明显升高,而旋转耐受时间降低(P<0.01);平肝潜阳法方药能够逆转这种改变。模型组AngⅡ和PDGF-BB含量及AT1αR表达较正常组显著升高(P<0.01,P<0.05),治疗组较模型组显著降低(P<0.05,P<0.01)。结论:平肝潜阳法方药不仅能够明显降低血压,而且能够改善大鼠的宏观表征,其作用可能与调节血管组织中AngⅡ和PDGF-BB表达有关。 Objective:To investigate the expression of AngⅡ and PDGF-BB in the vascular tissue of spontaneously hypertensive rats with the hyperactivity of liver-yang syndrome,and explore the action of Pinggan Qianyang Formula.Methods:Ten SD rats were used as the normal group,20 spontaneously hypertensive rats were randomly divided into the model group and the treatment group.A rat model with liver-yang hyperactivity was prepared with spontaneous hypertensive rats (SHRs) by administration of Aconiti Praeparatae Decoction.The rats in treatment group were administrated respectively with Pinggan Qianyang Formula for four weeks after reproduced the model.Monitoring the changes of behavior and blood pressure of rats were measured during experiments procedure,then they were taken their vascular tissue unified out at the seventh week.The level of AngⅡ and PDGF-BB were detected by enzyme-linked immunosorbent assay.qPCR and Western Blot were performed to detect the expression of AT1αR and PDGF-BB mRNA and protein in the vascular tissue.Results:The rat model of liver-yang hyperactivity was significantly higher,systolic blood pressure,grades of irritability and lower rotation endurance time than the SD rats (P<0.01).However,the changes were inversed by the application of Pinggan Qianyang Formula.The content of AngⅡ and PDGF-BB and the expression of AT1αR in the model group were significantly higher than those in the normal group (P< 0.01,P<0.05),and the content of AngⅡ and PDGF-BB and the expression of AT1αR were significantly lower in the treatment group than in the model group (P<0.05,P<0.01).Conclusion:The Pinggan Qianyang method can not only reduce blood pressure obviously,but also improve the macroscopic characterization of rats,which may be related to the regulation of the expression of AngⅡ and PDGF-BB in the vascular tissue.
