摘要
目的:观察当归四逆汤对动脉硬化闭塞症(ASO)寒凝血瘀型家兔血管平滑肌细胞(VSMC)中ERK1、c-myc mRNA表达的影响,探讨其防治ASO的作用机制。方法:以体外培养兔主动脉VSMC为研究对象,应用血清药理学法,采用Real-time PCR法检测兔VSMC中ERK1、c-myc mRNA的表达。结果:与正常组比较,模型组ERK1、c-myc mRNA表达水平显著升高(P<0.01);与模型组比较,各用药组ERK1、c-myc mRNA表达水平显著降低(P<0.01,P<0.05),其中当归四逆汤高剂量组ERK1、c-myc mRNA表达水平降低最为明显。结论:当归四逆汤通过抑制原位癌基因c-myc mRNA的表达和细胞信号通路ERK1 mRNA的表达,从而抑制了VSMC的增殖和迁移,阻止了ASO的发生和发展。
Obiective:To discuss the mechanism of Danggui Sini Decoction control athero sclerosis obliterans (ASO) by observing the effects of Danggui Sini Decoction on ERK1 mRNA,c-myc mRNA expression in vascular smooth muscle cells in rabbits of ASO cold blood coagulation and stasis.Methods:Rabbit aortic VSMC cultured in vitro as the research object,the application of serum pharmacological method,Real-time PCR method is used to detect the expression of ERK1 mRNA,c-myc mRNA in vascular smooth muscle cells.Results:The expression of ERK1 mRNA,c-myc mRNA increased significantly in AngⅡ group than in normal group (P<0.01) and reduced significantly in treated group (P<0.01,P<0.05).It reduced the most obvious in high-dose group.Conclusion:Danggui Sini Decoction control ASO by inhibiting the expression of ERK1 mRNA,c-myc mRNA and then inhibiting VSMC proliferation and migration.
