大补脾汤联合黄芪对顺铂肾脏毒性的干预作用

Effects of Dabupi Decoction combined with Astragalus on cisplatin nephrotoxicity

目的:探讨敦煌古方大补脾汤联合黄芪对顺铂肾脏毒性的干预作用。方法:60只昆明小鼠随机分为生理盐水组,模型组,顺铂+高、中、低剂量组(0.7、0.5、0.25mL),单独中药组(0.5mL)。除生理盐水组和单独中药组,其余小鼠腹腔注射顺铂造模后,进行大补脾汤联合黄芪中药干预。治疗30d后,测量各组小鼠的体质量、肾脏系数、血清中肌酐(Scr)和尿素氮(BUN),HE染色切片观察小鼠肾组织病理变化,应用实时荧光定量PCR(RT-PCR)方法检测小鼠肾脏组织TNF-α mRNA表达水平,酶联免疫吸附试验(ELISA)检测小鼠血清中TNF-α含量。结果:与模型组比较,顺铂+高剂量组小鼠的体质量、肾脏系数明显升高(P<0.01),血清中Scr、BUN、TNF-α含量均显著降低(P<0.01),肾脏组织炎性因子TNF-α mRNA表达水平下调(P<0.01),肾小球病变程度等均有所改善。结论:敦煌古方大补脾汤联合黄芪能一定程度改善顺铂的肾脏毒性。

Objective:To investigate the effects of Dabupi Decoction combined with Astragalus on cisplatin nephrotoxicity.Methods:Sixty Kunming mice were randomly divided into normal saline group,model group,Dabupi Decoction high,medium and low dose group (0.7,0.5,0.25mL),Chinese medicine control group (0.5mL).In addition to the normal saline group and the Chinese medicine control group,the other mice were injected with cisplatin intraperitoneally to make the model.After 30 days of treatment,the body weight,renal coefficient,serum creatinine (Scr) and urea nitrogen (BUN) of each group were measured.The pathological changes of renal tissue were observed by HE staining.The expression level of TNF-α mRNA in renal tissue was detected by Real-time PCR (RT-PCR),and the content of TNF-α in serum was detected by enzyme-linked immunosorbent assay (ELISA).Results:Compared with the normal?saline group,the body weight and kidney coefficient in Dabupi Decoction high-dose group were increased (P<0.01),the expression of Scr,BUN,TNF-α in serum were decreased (P<0.01),the mRNA expression of TNF-α in renal tissue was decreased (P<0.01),and alleviate renal?pathological?changes.Conclusion:Dabupi Decoction combined with Astragalus can improve the nephrotoxicity of cisplatin to some extent.
