小青龙汤对变应性鼻炎小鼠树突状细胞的影响 被引量:13

摘要 目的探讨小青龙汤治疗变应性鼻炎过程中对树突状细胞(DCs)免疫功能的影响。方法选取SPF级BALB/c小鼠30只,通过随机数字表法将其分为对照组、鼻炎模型组、小青龙汤组。利用卵清蛋白和氢氧化铝建立变应性鼻炎模型,观察小鼠在小青龙汤治疗后挠鼻和喷嚏次数的改变,苏木精-依红染色(HE染色)观察小鼠鼻黏膜病理变化。流式细胞仪检测小鼠肺泡灌洗液中DCs表面分子CD40、CD80、CD86数量。酶联免疫吸附试验检测小鼠血清IgE的浓度。结果鼻炎组小鼠挠鼻及喷嚏次数明显高于对照组(P<0.01),小青龙汤组明显低于鼻炎组(P<0.01)。与对照组相比较,模型组小鼠鼻甲黏膜肿胀,嗜酸性粒细胞浸润。小青龙汤组DCs数目及活化DCs表明分子CD40、CD80、CD86与模型组相比均呈不同程度降低(P<0.01)。模型组小鼠血清IgE浓度明显高于空白对照组,小青龙汤组低于模型组(P<0.05)。结论小青龙汤可以有效改善变应性鼻炎小鼠鼻部症状且可以有效抑制DCs活化和成熟,调节免疫功能降低炎症反应,从而起到治疗变应性鼻炎的作用。 Objective To determine the effect of Xiaoqinglong decoction on the immune function of dendritic cells(DCs) in the treatment of allergic rhinitis(AR). Methods Thirty SPF BALB/c mice were selected and divided into a control group, a rhinitis model group and a Xiaoqinglong decoction group by random number table method. The AR model was established by ovalbumin and aluminum hydroxide, and the changes of frequency of scratching and sneezing were observed in mice after the treatment with Xiaoqinglong decoction, and the pathological changes of nasal mucosa in mice were observed by hematoxylin-yihong staining(HE staining). Flow cytometry detected the number of CD40, CD80 and CD86 molecules on the surface of DCs cells in the bronchoalveolar lavage fluid of mice. Enzyme-linked immunosorbent assay was used to detect IgE concentration in the serum of mice. Results The frequency of scratching nose and sneezing in the rhinitis group was significantly higher than that in the control group(P < 0.01), while that in the Xiaoqinglong decoction group was significantly lower than that in the rhinitis group(P < 0.01). Compared with the control group, the model group showed turbinate mucosa swelling and eosinophil infiltration. The number of DCs and activated DCs cells in the Xiaoqinglong decoction group showed that the molecular CD40, CD80 and CD86 were decreased to different degrees compared with the model group(P < 0.01). The serum IgE concentration of the model group was significantly higher than that of the control group, while that of the Xiaoqinglong group was lower than that of the model group(P < 0.05). Conclusion Xiaoqinglong decoction effectively improves the nasal symptoms in mice with allergic rhinitis, inhibits the activation and maturation of DCs, regulates the immune function and reduces the inflammatory response.
