桦褐孔菌总多糖抗糖尿病作用的研究 被引量:3

摘要 目的研究桦褐孔菌总多糖(IOTP)对链脲佐菌素(STZ)诱导的糖尿病大鼠的抗糖尿病作用.方法利用水提醇沉法提取桦褐孔菌总多糖(IOTP).50只雄性SD大鼠随机分为正常对照组、模型组、阳性对照组(800μg/kg)、IOTP 400 mg/kg组和IOTP 800 mg/kg组,每组10只.采用60 mg/kg STZ柠檬酸钠缓冲溶液单次腹腔注射造模.造模1周后,血糖值高于11.1 mmol/L的大鼠被认为是糖尿病大鼠.给药14 d后,测定其血糖、胰岛素水平、血脂(TC、TG、LDL-C和HDL-C)、ALT、AST、LDH活性.HE染色观察胰腺的病理学改变.结果与正常对照组相比,模型组大鼠的血胰岛素、HDL-C水平显著下降(P<0.01),血糖值、TC、TG、LDL-C、AST、ALT和LDH水平显著升高(P<0.01).与模型组相比,IOTP 800 mg/kg组和阳性对照组大鼠血胰岛素、HDL-C水平显著升高(P<0.01),血糖、TC、TG、LDL-C、AST、ALT和LDH水平显著降低(P<0.01);IOTP 800 mg/kg组胰岛萎缩和胰腺细胞空泡化程度被降低.结论IOTP对STZ诱导的糖尿病大鼠不仅具有显著降糖作用,而且能够调节血脂、抑制胰腺和肝脏的损伤. Objective To investigate the antidiabetic effects of Inonotus obliquus total polysaccharides(IOTP)in streptozotocin(STZ)-induced diabetic rats.Methods IOTP was extracted with water extraction followed by alcohol precipitation.Fifty male SD rats were randomly divided into normal group,model group,positive control group(glibenclamide 800μg/kg),IOTP 400 mg/kg group,and IOTP 800 mg/kg group,with 10 rats in each group.The diabetic model was induced by single intraperitoneal injection of 60 mg/kg STZ-sodium citrate buffer solution.The rats with blood glucose levels above 11.1 mmol/L were considered diabetic rats after one week of modeling.After the administration of IOTP for 14 d,the levels of blood glucose,insulin,blood lipids(total cholesterol,triglyceride,LDL-C and HDL-C),and the activities of alanine aminotransferase(ALT),aspartate aminotransferase(AST),lactate dehydrogenase(LDH)were detected.Meanwhile the pathological changes of pancreatic were detected by HE staining.Results Compared with normal control group,the blood insulin and HDL-C levels were significantly decreased in model group(P<0.01),and the blood glucose levels,TC,TG,LDL-C,AST,ALT,and LDH levels were significantly increased(P<0.01).Compared with model group,the blood insulin and HDL-C levels of rats were significantly increased in IOTP 800 mg/kg group and positive control group(P<0.01),while the blood glucose,TC,TG,and LDL-C,AST,ALT and LDH levels were decreased(P<0.01).The islet atrophy and vacuolization of pancreatic cells in IOTP 800 mg/kg group were reduced.Conclusion IOTP not only has significant anti-diabetic effects in STZ-induced diabetic rats,but also can regulate blood lipids and inhibit pancreatic and liver damage.
