肺纤方对博来霉素诱导的肺间质纤维化大鼠上皮间质转化的影响 被引量:4

Effects of Feixian Formula on epithelial-mesenchymal transition in bleomycin-induced pulmonary interstitial fibrosis in rats

摘要 目的:通过观察肺纤方对博来霉素诱导的肺间质纤维化大鼠肺组织病理学的改变以及对α-SMA、E-cadherin及Ⅰ型胶原的mRNA表达的影响,探讨肺纤方抗肺间质纤维化的作用机制。方法:将120只Wistar大鼠随机分为假手术组,模型组,泼尼松组,肺纤方高、中、低剂量组,每组20只。除假手术组外,其余组均采用气管插管滴注博来霉素3mg/kg进行肺间质纤维化大鼠造模。造模第2天进行药物灌胃干预,并于第14、28天分两批处死动物,取出肺脏,HE和Masson染色分别观察肺泡炎和肺间质纤维化的程度;Real time PCR检测α-SMA、E-cadherin、Ⅰ型胶原的mRNA表达。结果:肺纤方高、中剂量可抑制肺泡炎和肺纤维化;第14、28天,与模型组比较,肺纤方高剂量组能显著降低肺组织中α-SMA、Ⅰ型胶原的mRNA表达(P<0.01,P<0.05),增加E-cadherin的mRNA表达(P<0.01,P<0.05)。结论:肺纤方通过抑制上皮间质转化而发挥抗肺间质纤维化的作用。 Objective:To investigate the influence of Feixian Formula(FXF)on pulmonary histopathology and the expression ofα-SMA,E-cadherin and CollagenⅠin bleomycin(BLM)-induced pulmonary fibrosis in rats and explore its potential therapeutic mechanisms.Methods:A total of 120 Wistar rats were randomly divided into control group,BLM group,BLM+Prednisone(Pred)group,BLM+FXF high,middle and low dose groups,20 rats in each group.3mg/kg BLM was injected intratracheally to induce pulmonary fibrosis model except control group.The rats were killed on day 14 and 28 respectively.The lung tissues were collected to make HE staining and Masson staining,to observe the degree of alveolitis and pulmonary interstitial fibrosis.The lung tissues were also collected to detect the expression ofα-SMA,E-cadherin and CollagenⅠby Real time PCR.Results:The high and middle dose of FXF could inhibit alveolitis and pulmonary fibrosis.On day 14 and 28,compared with the model group,the BLM+FXF high dose group could significantly reduce the mRNA expression ofα-SMA and CollagenⅠ(P<0.01,P<0.05)and increase the mRNA expression of E-cadherin(P<0.01,P<0.05)in the lung tissues.Conclusion:The protective role of FXF in pulmonary fibrosis rats is mediated by suppressing epithelial-mesenchymal transition.
