苹果轮纹病抗性候选基因的表达分析 被引量:1

摘要 轮纹病菌侵染苹果植株,使树体减弱,严重影响苹果产量和品质。研究苹果轮纹病抗性候选基因的表达分析,以期为揭示苹果轮纹病的抗病机制提供参考。本研究以河北农业大学苹果课题组筛选出的抗病株系‘1-1-46’和感病株系‘1-1-40’为试材,对枝条接种轮纹病菌(B.berengeriana f.sp.piricola)和空白培养基,观察0~40 d内枝皮中的轮纹病抗性候选基因的表达量。结果表明:接种轮纹病菌后感病株系‘1-1-40’可以观察到明显的病变现象,皮孔凸起呈瘤状,抗病株系‘1-1-46’无明显的变化。对枝皮进行抗病候选基因Mdcf3、MdMBP2(苹果甘露糖结合蛋白)和MdACBP2(苹果酰基辅酶A结合蛋白)的表达分析,结果表明,接种轮纹病菌后,Mdcf3基因的相对表达量感病株系‘1-1-40’低于抗病株系‘1-1-46’,MdACBP2基因和MdMBP2基因相对表达量抗病株系‘1-1-46’低于感病株系‘1-1-40’。由此认为,对一些试材测定基因的相对表达量可以作为植株抗、感性鉴定的参考评价指标。 The ringworm infects the apple plants,which weakens the tree and seriously affects the yield and quality of the apple.Improving the disease resistance of apples by means of molecular breeding provides a favorable theoretical basis for subsequent research.In this study,the disease-resistant strains'1-1-46'and susceptible strains'1-1-40'selected by the Apple Research Group of Agricultural University of Hebei were used as experiment materials,and the branches were infected by B.berengeriana f.sp.piricola and blank medium were used to observe the expression level of the candidate for ringworm resistance in the infection within 0~40 days.The results showed that obvious pathological changes were observed in the susceptible strain'1-1-40'after infected by B.berengeriana f.sp.piricola,and the lenticulus was tumor-like,and the resistant strain'1-1-46'had no obvious change.The scalp can be slightly raised by hand.The expressions of the disease resistance candidate genes Mdcf3,MdMBP2 and MdACBP2 in the branches were analyzed.The results showed that the relative expression of Mdcf3 gene,'1-1-40'was lower than the resistance after infected by B.berengeriana f.sp.piricola.The diseased strain'1-1-46',the relative expression of MdACBP2 gene and MdMBP2 gene resistant strain'1-1-46'was lower than the susceptible strain'1-1-40'.It is indicated that the relative expression level of some tested materials can be used as a reference evaluation index for plant resistance and sensitivity identification.
