槲皮素联合喜树碱促进人前列腺癌PC-3细胞的凋亡

Pro-apoptotic effects of quercetin combined with camptothecin on human prostate cancer PC-3 cell

目的考察槲皮素联用喜树碱对人前列腺癌PC-3细胞凋亡的影响。方法体外培养PC-3细胞,用CCK-8实验、细胞周期检测实验和Annexin V-FITC/PI双染细胞凋亡检测实验,考察槲皮素联用喜树碱对PC-3细胞增殖凋亡的影响;用Western-Blot实验检测凋亡相关蛋白(Bcl-2、Bax、PARP1和Caspase-3)的表达。结果 CCK-8实验结果显示槲皮素和喜树碱都可以一定程度地抑制PC-3细胞增殖,诱导凋亡作用,其中槲皮素和喜树碱联用效果更优。药物作用48 h后其半数抑制浓度(IC_(50))分别为38.76、2.25、1.88μmol·L^(-1)。细胞周期实验显示,药物联用组细胞周期处于S期的细胞比例明显高于单用组(P<0.05);细胞凋亡实验显示,药物联用组的细胞凋亡率显著高于单用组(P<0.05);Western-Blot结果显示,药物联用组对凋亡相关蛋白的影响更为明显,使Bcl-2蛋白表达下降为47%,分别使Bax、PARP1和Caspase-3依次增加1.17、0.77和0.69倍(P<0.05)。结论槲皮素联合喜树碱能够显著促进人前列腺癌PC-3细胞的凋亡。

AIM To investigate the effect of quercetin combined with camptothecin on apoptosis of human prostate cancer PC-3 cells.METHODS Human prostate cancer PC-3 cells were cultured in vitro.The apoptosis effects of drugs on PC-3 cells were determined by CCK-8 assay,Annexin V-FITC/PI bistaining and cell cycle analysis.Western-Blot was used to investigate the expression of apoptosis-related proteins(Bcl-2,Bax,PARP1 and Caspase-3)and to explore its mechanism.RESULTS CCK-8 results showed that both quercetin and camptothecin could inhibit the proliferation of PC-3 cells and induce apoptosis.Among them,the combination of two drugs was more effective,and the IC50 was 38.76,2.25 and 1.88μmol·L-1 after 48 h of drug intervention,respectively;the cell cycle analysis showed that the proportion of cells at S phase in the drug combination group was significantly higher than that in the single groups(P<0.05).The Annexin V-FITC/PI bistaining showed that the apoptosis rate of PC-3 cells in the combination group was significantly higher than that in the single groups(P<0.05).Western-Blot showed that the effect of combination group on apoptosis-related proteins was more obvious,Bcl-2 decreased to 47%,and Bax,PARP1 and Caspase-3 increased by 1.17,0.77 and 0.69 times,respectively.CONCLUSION Quercetin combined with camptothecin can significantly promote the apoptosis of human prostate cancer PC-3 cells.
