摘要
为了探究旋毛虫感染诱导的宿主免疫对H22荷瘤小鼠肿瘤生长的影响,试验随机将100只Balb/c小鼠分成空白组、旋毛虫组、荷瘤组、旋毛虫+荷瘤组,每组25只。旋毛虫+荷瘤组小鼠在感染旋毛虫后的第7天(试验第7天)接种小鼠肝癌H22细胞,荷瘤组与旋毛虫+荷瘤组小鼠接种肝癌H22细胞的时间相同。在试验第28天,荷瘤组和旋毛虫+荷瘤组均处死10只小鼠用于计算抑瘤率。在试验的第7,14,21,28,35天每组各处死3只小鼠并收集脾细胞进行培养,采用ELISA法和实时荧光定量PCR技术对各组小鼠脾脏中白细胞介素-2(IL-2)、干扰素-γ(IFN-γ)和白细胞介素-4(IL-4)的表达量进行检测。结果表明:与荷瘤组相比,旋毛虫+荷瘤组的抑瘤率为62.65%(P<0.05),旋毛虫感染有效抑制了H22肿瘤的生长。旋毛虫+荷瘤组IL-2、IFN-γ的表达量于第7,14,21天明显升高,与空白组和荷瘤组相比差异显著(P<0.05);旋毛虫+荷瘤组IL-4的表达量总体呈逐渐升高趋势。说明旋毛虫感染早期诱导宿主产生Th1细胞因子IL-2、IFN-γ以及后期诱导宿主产生Th2细胞因子IL-4可能抑制肿瘤的进一步发展。
To investigate the effect of host immunity induced by Trichinella spiralis(T.spiralis)infection on tumor growth in H22 tumor-bearing mice,100 Balb/c mice were randomly divided into control group,T.spiralis group,tumor group,and T.spiralis+tumor group,25 in each group.The mice of the T.spiralis+tumor group were inoculated with H22 hepatocellular carcinoma cells on the 7th day after infection with T.spiralis.The mice of the tumor group and the T.spiralis+tumor group were inoculated with H22 hepatocellular carcinoma cells at the same time point.On the 28th day of the experiment,10 mice in each of the tumor group and the T.spiralis+tumor group were sacrificed to calculate the tumor inhibition rate.On 7th,14th,21st,28th and 35th days of the experiment,3 mice of each group were sacrificed and spleen cells were collected for culture.ELISA and Quantitative Real-time PCR were used to detect the changes of interleukin-2(IL-2),interferon-γ(IFN-γ)and interleukin-4(IL-4)expression in each group.The results showed that compared with the tumor group,the tumor inhibition rate of the T.spiralis+tumor group was 62.65%(P<0.05),T.spiralis infection effectively inhibited the growth of H22 tumors.The expression levels of IL-2 and IFN-γin T.spiralis+tumor group increased significantly on the 7th,14th and 21st days,which were significantly different from those in the control group and tumor group(P<0.05).The expression of IL-4 in the T.spiralis+tumor group showed an overall increasing trend.The results of this study indicated that IL-2 and IFN-γof the Th1-type cytokines in the early phase of T.spiralis infection and IL-4 of Th2-type cytokines anti-inflammation cytokine in the late phase of T.spiralis infection might contribute to the inhibition of tumor growth.
