摘要
目的:研究参麻益智方对血管性痴呆(VD)模型大鼠认知功能和脑白质损伤的影响。方法:将60只Wistar大鼠经造模手术后随机分为假手术组、模型组、参麻益智方低剂量组、参麻益智方中剂量组、参麻益智方高剂量组和多奈哌齐组。给药2周后行水迷宫实验检测大鼠的认知功能,脑组织LFB染色观察胼胝体髓鞘脱失情况并进行脑白质损伤评分,Western blot法检测MBP蛋白表达。结果:参麻益智方高剂量组大鼠水迷宫逃避潜伏期明显缩短,目标象限停留时间明显延长,脑白质损伤评分明显降低,MBP表达水平明显升高。结论:参麻益智方可以改善VD大鼠的认知功能,减轻VD大鼠的脑白质损伤。
Objective:To study the effect of Shenma Yizhi Decoction(SMYZD)on cognitive function and white matter lesions in VD rats.Methods:A total of 60 surgery modeled Wistar rats were randomly divided into Sham group,Model group,SMYZD-L group,SMYZD-M group,SMYZD-H group and Donepezil group.After 2 weeks administration of SMYZD,Morris water maze test was used to detect cognitive function of rats.Brain tissue LFB staining was used to observe loss of myelin sheath of the Corpus callosum and White matter damage scoring.Western blot was used to detect the expression of myelin basic protein(MBP).Results:Escape latency of rats in SMYZD-H group significantly shortened.Time spent in target quadrant significantly prolonged.Grading score of white matter damage significantly diminished,and MBP expression augmented.Conclusion:Shenma Yizhi Decoction could improve cognitive dysfunction and alleviate white matter damage of VD rats.
