聚乳酸/海藻多糖电纺支架的制备工艺研究

摘要 目的:通过夹层静电纺丝法,制备具有良好生物相容性的聚乳酸纤维-明胶/多糖纤维的夹层复合小口径组织工程血管支架。方法:将不同浓度的聚乳酸、海藻多糖明胶溶液,通过高压静电场作用进行静电纺丝,并对不同多糖浓度的电纺支架进行力学性能及血小板粘附性的测定。结果:随着海藻多糖浓度的增加,电纺支架抗拉强度逐渐增大,断裂伸长率基本不变,爆破强度接近人体血管所承受的最大压力,血小板黏附性逐渐减弱。结论:聚乳酸浓度为10%、接收距离为12 cm、电压为12 kV、纺丝液流速为0.8 mL/h,海藻多糖明胶溶液静电纺丝接受距离为10 cm、电压为15 kV、流速为4.8 mL/h时,电纺支架性能最优。 objective:to prepare poly(lactic acid)fiber-gelatin/polysaccharide composite scaffolds with good biocompatibility by interlayer electrospinning.Methods:the polylactic acid and seaweed polysaccharide gelatin solutions with different concentrations were electrospun by the action of high voltage electrostatic field,and the mechanical properties and platelet adhesion of the electrospun scaffolds with different polysaccharide concentrations were determined.Results:with the increase of seaweed polysaccharide concentration,the tensile strength of electrospinning scaffold gradually increased,the elongation at break was basically unchanged,the blasting strength was close to the maximum pressure of human blood vessels,and the adhesion of platelets gradually weakened.Conclusion:when the polylactic acid concentration is 10%,the receiving distance is 12 cm,the voltage is 12 kV,the spinning fluid flow rate is 0.8 mL/h,and the receiving distance of seaweed polysaccharide gelatin solution is 10 cm,the voltage is 15 kV,and the flow rate is 0.08 mL/min,the electrospinning scaffold has the best performance.
