响应面分析法优化吴茱萸中吴茱萸碱和吴茱萸次碱的超声提取工艺 被引量:1

Optimization of Ultrasonic Extraction of Evodiamine and Rutaecarpine from Evodia Rutaecarpa by Response Surface Methodology

摘要 以吴茱萸碱和吴茱萸次碱提取率之和为考察指标,采用HPLC法检测其含量。首先考察溶剂、目数、液固比、提取温度、超声功率和提取时间等单因素对提取率的影响;再通过响应面分析法对吴茱萸中吴茱萸碱和吴茱萸次碱的提取工艺进行优化。结果表明,超声提取吴茱萸中生物碱的最佳工艺条件为:溶剂为甲醇、目数为20~40目、超声功率为420 W、提取温度为51℃、提取时间为52 min、液固比为51∶1(mL/g),提取率的预测值为2.529 mg/g,验证值为2.456 mg/g,与预测值的相对误差为2.88%。 Taking the sum of the extraction rate of evodiamine(Evo)and rutaecarpine(Rut)as the index,using high performance liquid chromatography(HPLC)detected the content.Firstly,we investigated the influence of the single factor:the solvent,the mesh number,the liquid-solid compare,the extraction temperature,the extraction power and the extraction time for the single factor experiments,the extraction temperature,the extraction time and the liquid-solid compare for the extraction rate;then suing the Response Surface Method(RSM)to optimize the the extract condition of evodiamine and rutaecarpine.Result showed,the optimum conditions of evodiamine and rutacarpine extraction were:solvent methanol,material mesh 20~40,ultrasonic power 420 W,extraction temperature 51℃,extraction time 52 min,liquid-solid compare 51∶1(mL/g).Under these conditions,the yield of the Evo&Rut was 2.456 mg/g,which differed from the predicted values of 2.529 mg/g only by the relative error of 2.88%.
