摘要
目的:探讨沙门氏菌烈性噬菌体的分离鉴定与生物学特性。方法:使用双层平板法从临病死鸡的粪便及垫料中分离沙门氏菌噬菌体CR5,对分离到的噬菌体实施纯化、裂解谱检测、效价检测和电镜形态观察,并进行噬菌体温度稳定性试验,了解温度变化对噬菌体产生的影响。结果:噬菌体CR5蚀斑均匀、透亮,效价为109 PFU/mL;经裂解谱检测发现CR5可对多株沙门氏菌菌株进行裂解,属于宽裂解谱噬菌体;电镜观察结果提示,CR5呈长尾状态;CR5在40℃、50℃和60℃温度条件下可维持较好活性,在70℃条件下会出现效价快速下降情况,80℃温度下20 min后即可失活,温度耐受性良好。结论:噬菌体CR5是一种长尾科裂解性噬菌体,具有良好的抑菌效用,温度耐受度较好。
Purpose:To investigate the isolation,identification and biological characteristics of Salmonella phage.Method:The double-layer plate method was used to isolate Salmonella phage CR5 from the feces and litter of dead chickens,and the isolated phages were purified,lysed by spectroscopy,titered,and observed under electron microscopy.The effect of changes on phage production.Result:The phage CR5 plaque is uniform and translucent with a potency of 109 PFU/mL;CR5 can lyse multiple Salmonella strains by lysis spectrum detection,and belongs to a wide lysis spectrum phage.Electron microscope observation results indicate that CR5 is in a long tail state;It can maintain good activity under the conditions of 40℃,50℃and 60℃,and the titer will decrease rapidly at 70℃.After 80 minutes at 80℃,it will be inactivated and have good temperature tolerance.Conclusion:The phage CR5 is a long-tailed lysing phage,which has good bacteriostatic effect and good temperature tolerance.
