摘要
目的观察Meprinα对转化生长因子(TGF)-β1介导的成纤维细胞胶原合成和肌成纤维细胞分化的调节作用。方法原代培养肺成纤维细胞,采用TGF-β1诱导,并予以重组Meprinα及放线酰胺素(Actinonin)预处理。免疫细胞化学染色检测α-平滑肌肌动蛋白(α-SMA)的表达,免疫印迹法检测Meprinα、I型胶原、α-SMA的表达。结果 TGF-β1能够促进肺成纤维增殖和迁移能力,显著上调I型胶原、α-SMA的表达,同时降低Meprinα的表达(P<0.05)。免疫印迹结果显示,予以Meprinα预处理能够显著抑制TGF-β1介导的I型胶原、α-SMA表达的上调(P<0.05);予以Actinonin预处理,则能够显著促进TGF-β1介导的I型胶原、α-SMA表达的上调(P<0.05)。结论 Meprinα具有抑制TGF-β1介导的肺成纤维细胞胶原合成和肌成纤维细胞分化的作用。
Objective To explore the role of meprin α on collagen synthesis and myofibroblast differentiation in fibroblasts induced by TGF-β1. Me thods Primary lung fibroblasts were cultured and induced by TGF-β1,and treated with meprin α and actinonin. The expression of α-SMA was observed by immunofluorescent staining.The expressions of meprin α, collagen type I(COL I), and α-SMA were measured by Western blot. Re s ults TGF-β1 promoted cell proliferation and migration in lung fibroblasts, accompanied with an increase of COL I andα-SMA, and a decrease of meprin α (P<0.05). Pretreatment with meprin α could inhibite the levels of COL I andα-SMA induced by TGF-β1(P<0.05). And pretreatment with actinonin promoted the levels of COL I andα-SMA induced by TGF-β1(P <0.05). Conclus ion Meprinα can inhibit collagen synthesis and myofibroblast differentiation in lung fibroblasts induced by TGF-β1.
