氢吗啡酮后处理对大鼠心肌缺血再灌注损伤的保护作用及机制 被引量:5

Protective effect and mechanism of hydromorphine post-processing for myocardial ischemia-reperfusion injury in rats

摘要 目的探究氢吗啡酮后处理对大鼠心肌缺血再灌注损伤的保护作用及机制。方法将54只健康雄性SD大鼠随机分为假手术组、模型组及实验组,各18只。模型组及实验组大鼠采用结扎左冠状动脉前支构建心肌缺血-再灌注损伤大鼠模型;假手术组则仅穿线不结扎。实验组及模型组于结扎25 min后,尾静脉分别注射0.3μmol·L^-1氢吗啡酮注射液及等量0.9%氯化钠,缺血30 min后再灌注120 min,假手术组予以0.9%氯化钠。用硫代巴比妥酸法检测血清丙二醛(MDA)含量,用羟胺法检测超氧化物歧化酶(SOD)含量,用TUNEL法检测心肌细胞凋亡情况。结果假手术组、模型组及实验组再灌注结束后血清MDA含量分别为(6.14±2.05),(23.07±4.17),(12.45±3.09)nmol·mL^-1;SOD含量分别为(52.31±6.89),(19.79±4.02),(29.56±4.61)U·mL^-1;心肌细胞凋亡指数分别为(1.02±0.75)%,(26.87±3.82)%,(13.21±2.11)%;模型组的上述指标分别与假手术组、实验组比较,差异均有统计学意义(均P<0.05)。结论氢吗啡酮后处理有效抑制心肌缺血再灌注损伤大鼠氧化应激反应,缓解心肌损伤,其作用机制与抑制心肌细胞凋亡相关。 Objective To explore the protective effect and mechanism of hydromorphine post-processing for myocardial ischemia-reperfusion injury in rats.Methods A total of 54 healthy male SD rats were randomly assigned to sham-operation group,model group and test group,18 rats were in each group.Myocardial ischemia-reperfusion injury rat model was built in the model group and the test group by ligation of the left anterior coronary artery,while the sham-operation group received no ligation.The test group and the model group treated with tail intravenous injection for 0.3μmol·L^-1 of hydromorphine injection and equal amount of 0.9%NaCl for 25 min after ligation respectively,30 min after ischemia reperfusion for 120 min.The sham-operation group was given 0.9%NaCl.The content of serum malondialdehyde(MDA)after reperfusion was detected by glucosinolates barbituric acid method,and the content of superoxide dismutase(SOD)was detected by hydroxylamine method;the myocardial cell apoptosis was detected by TUNEL me-thod.Results The serum MDA contents in sham-operation group,model group and test group after reperfusion were(6.14±2.05),(23.07±4.17),(12.45±3.09)nmol·m L^-1,respectively,the SOD contents were(52.31±6.89),(19.79±4.02),(29.56±4.61)U·m L^-1,respectively;the myocardial cell apoptosis indexes were(1.02±0.75)%,(26.87±3.82)%,(13.21±2.11)%,respectively.The above indexes in model group had statistically significant difference compared with those in sham-operation group,test group(all P<0.05).Conclusion Hydromorphine post-processing can inhibit the oxidative stress reaction,alleviate myocardial injury,which might be associated with inhibition of myocardial cell apoptosis.
