吴茱萸碱对淋巴瘤移植小鼠模型肿瘤组织Ki-67表达的影响 被引量:3

Effect of evodiamine on the Ki-67 expression in tumor tissue of lymphoma transplanted mice model

摘要 目的研究吴茱萸碱对淋巴瘤移植小鼠模型肿瘤组织肿瘤细胞增殖核抗原(Ki-67)表达的影响。方法选取45只建模成功的淋巴瘤移植模型小鼠,随机分为模型组及低、高剂量实验组,各15只。建模成功后第1,3,5天,低、高剂量实验组灌胃10,20 mg·kg^-1·d^-1吴茱萸碱,模型组则灌胃等量生理盐水。分别于干预后1,2,3周比较各组小鼠体重、肿瘤体积变化;以逆转录-聚合酶链反应(RT-PCR)法和免疫组化法检测干预后3周各组肿瘤组织Ki-67 mRNA及Ki-67蛋白表达情况。结果模型组及低、高剂量实验组小鼠干预后3周肿瘤体积分别为(396.14±10.95),(239.57±10.01),(191.01±9.79)mm^3,肿瘤组织中Ki-67 mRNA相对表达量分为0.54±0.09,0.33±0.10,0.27±0.08,Ki-67蛋白免疫组化评分分别为(8.23±0.87),(5.91±0.96),(4.03±0.84)分。干预后1~3周,模型组及低、高剂量实验组小鼠体重及肿瘤体积逐渐增大,且不同时间点低、高剂量实验组较模型组降低(P<0.05);与模型组比较,低、高剂量实验组小鼠肿瘤组织中Ki-67 mRNA及Ki-67蛋白水平显著降低,且高剂量实验组低于低剂量实验组(P<0.05)。结论吴茱萸碱可有效抑制淋巴瘤抑制小鼠肿瘤生长,其作用机制可显著下调肿瘤组织Ki-67表达相关。 Objective To explore the effect of evodiamine on the Ki-67 expression in tumor tissue of lymphoma transplanted mice model.Methods Forty-five SPF KM mice were built lymphoma transplanted mice model and randomly divided into model group and experimental-L/H groups,15 cases in each group.On 1,3,5 d after successfully modeling,the experimental-L/H groups were gavaged with 10,20 mg·kg^-1·d^-1 evodiamine,model group was intragastric administrated equal amount of normal saline.The mice body weight and tumor volume changes in each group at 1,2,3 weeks after intervention were compared;the Ki-67 mRNA and Ki-67 protein expression in tumor tissue 3 weeks after intervention were detected by reverse transcription polyme-rase chain reaction(RT-PCR)and immunohistochemistry.Results The tumor volumes at 3 weeks after intervention in model group and experimental-L/H groups were(396.14±10.95),(239.57±10.01),(191.01±9.79)mm^3,Ki-67 mRNA relative expression in tumor tissue were 0.54±0.09,0.33±0.10,0.27±0.08,Ki-67 protein immunohistochemistry scores were(8.23±0.87),(5.91±0.96),(4.03±0.84)scores.From 1 to 3 weeks after intervention,the mice body weight in model group and experimental-L/H groups increased gradually,tumor volume shrinked(P<0.05);the Ki-67 mRNA relative expression in tumor tissue and Ki-67 protein immunohistochemistry scores in experimental-L/H groups were lower than those in model group significantly,and experimental-H group was lower than experimental-L group(P<0.05).Conclusion Evodiamine can inbit the tumor growth of lymphoma transplanted mice effectively,the action mechanism may be related to the down-regulation of Ki-67 expression in tumor tissue significantly.
