白细胞介素-17A抗体对七氟烷麻醉的老年大鼠认知功能的保护作用及机制研究

摘要 目的探讨白细胞介素-17A(IL-17A)抗体对七氟烷麻醉的老年大鼠认知功能的保护作用及机制研究。方法将36只SD大鼠随机分为空白组、对照组和实验组,每组12只。空白组于侧脑室给予0.9%NaCl 3μL后连续吸入氧气和氮气(1∶1)混合气体4 h;对照组于侧脑室给予0.9%NaCl 3μL后连续吸入3%七氟烷4 h,实验组于侧脑室给予200μg·μL^-1 IL-17A抗体3μL后连续吸入3%七氟烷4 h。用Morris水迷宫实验记录逃避潜伏期评估大鼠认知功能,用酶联免疫检测试剂盒测定IL-17A与C反应蛋白(CRP)表达情况,用Western Blot法检测海马组织中Caspase-3、BACE-1和β淀粉样蛋白(Aβ)的表达水平。结果实验组、对照组和空白组的第5天逃避潜伏期分别为(26.33±3.98),(46.17±4.17)和(12.50±3.39)s,IL-17A分别为(102.24±11.16),(111.27±18.71)和(81.73±13.22)pg·mL^-1,CRP分别为(4.98±0.50),(5.49±0.32)和(4.75±0.54)pg·mL^-1,Caspase-3蛋白分别为(0.43±0.04),(0.89±0.04)和(0.37±0.05),BACE-1蛋白分别为(0.45±0.02),(0.97±0.06)和(0.41±0.08),Aβ蛋白分别为(0.34±0.07),(0.92±0.03)和(0.31±0.06),对照组的上述指标与空白组和实验组比较,差异均有统计学意义(均P<0.05)。结论IL-17A抗体可明显改善七氟烷麻醉的老年大鼠认知功能,降低炎性反应,抑制海马组织中Caspase-3、BACE-1和Aβ的表达。 Objective To investigate the protective effect and mechanism of interleukin-17 A(IL-17 A)antibody on cognitive function in elderly rats under sevoflurane anesthesia.Methods 36 SD rats were randomly divided into blank group,control group and experimental group,12 in each group.Rats in the blank group were given 0.9%NaCl 3μL in the lateral ventricle and continuously inhaled oxygen and nitrogen(1∶1)mixed gas for 4 h.Rats in the control group were given 0.9%NaCl 3μL and continuously inhaled 3%sevoflurane for 4 h.Rats in the experimental group were given 200μg·μL^-1 IL-17 A antibody 3μL and then continuously inhaled 3%sevoflurane for 4 h.Cognitive function in rats was evaluated by Morris water maze test to record escape latency.Expressions of IL-17 A and C-reactive protein(CRP)were determined by enzyme-linked immunosorbent assay kit.The expressions of Caspase-3,BACE-1 and beta-amyloid protein(Aβ)in the hippocampus were detected by Western Blot.Results The escape latencies on the 5 th day of the experimental,control and blank groups were(26.33±3.98),(46.17±4.17)and(12.50±3.39)s,IL-17 A were(102.24±11.16),(111.27±18.71)and(81.73±13.22)pg·m L^-1,CRP were(4.98±0.50),(5.49±0.32)and(4.75±0.54)pg·m L^-1,Caspase-3 protein were(0.43±0.04),(0.89±0.04)and(0.37±0.05),BACE-1 protein were(0.45±0.02),(0.97±0.06)and(0.41±0.08),and Aβprotein were(0.34±0.07),(0.92±0.03)and(0.31±0.06),respectively.The above-mentioned indexes in the control group were significantly different from those in blank group and experimental group(all P<0.05).Conclusion IL-17 A can significantly improve the cognitive function of elderly rats under sevoflurane anesthesia,reduce the inflammatory response,and inhibit the expressions of Caspase-3,BACE-1 and Aβin the hippocampus.
