适合培育蚕蛹虫草的蓖麻蚕品种和蛹虫草菌株筛选试验 被引量:1

摘要 用4个蓖麻蚕品种的5龄4天幼虫和4株蛹虫草菌(Cordyceps militaris)菌株培育蚕蛹虫草,以蓖麻蚕感染僵化率、出草率以及蚕蛹虫草中腺苷、虫草素、喷司他丁含量为指标,筛选最佳培育蚕蛹虫草的蓖麻蚕品种和蛹虫草菌株组合。结果显示,相同品种的蓖麻蚕幼虫接种不同蛹虫草菌株后僵化率差异极显著,接种后240 h接种H71菌株的僵化率最高为94.33%;不同品种蓖麻蚕接种相同蛹虫草菌株的僵化率差异显著,接种后240 h B2品种的僵化率最高为88.00%;不同试验组均获得正常子实体,平均出草率变幅为80.75%~90.45%,其中H71菌株接种B2品种的出草率最高,为94.64%;各试验组间蚕蛹虫草的子实体和僵蚕中虫草素、喷死他丁、腺苷含量达到极显著或显著差异,其中H22菌株接种B2品种所产生的子实体中喷司他丁和虫草素含量最高,质量分数分别为8.07 mg/g和8.45 mg/g,H71菌株接种B21品种所产生的子实体中腺苷含量最高,质量分数为5.24 mg/g;不同试验组僵蚕中虫草素、喷死他丁和腺苷含量均极显著(P<0.01)低于蚕蛹虫草子实体中的含量,其中H22菌株接种B21品种试验组僵蚕中的喷司他丁、虫草素和腺苷含量最高,质量分数分别为0.39 mg/g、2.67 mg/g和1.10mg/g。综合以上试验结果,初步认为以H22菌株接种B2品种培育的蚕蛹虫草,具有较高的喷司他丁和虫草素药用生产前景。 The 4 th day 5 th instar larvae from 4 varieties of Philosamia cynthia ricini and 4 strains of Cordyceps militaris were selected to culture aweto.The most suitable combination of Philosamia cynthia ricini variety and Cordyceps militaris strain was screened out according to indicators like infection rate,fruiting rate,and contents of adenosine,cordycepin and pentostatin in aweto.The results showed that the same variety of Philosamia cynthia ricini inoculated with different strains of Cordyceps militaris had extremely significant difference in infection rate.The strain H71 had the highest infection rate(94.33%)at 240 h post inoculation.In contrast,different varieties of Philosamia cynthia ricini inoculated with the same strain of Cordyceps militaris had significant difference in infection rate.The variety B2 had the highest infection rate(88.00%)at 240 h post inoculation.Normal fruiting body was obtained in all testing groups.The amplitude of average fruiting rate was from 80.75%to 90.45%,and the highest fruiting rate of 94.64%was found in variety B2 inoculated with strain H71.Furthermore,there were extremely significant difference or significant difference in content of cordycepin,pentostatin and adenosine in fruiting body and stiff larva from different testing groups.Among them,the highest contents of cordycepin and pentostatin were found in fruiting body of variety B2 inoculated with strain H22,which were 8.07 mg/g and 8.45 mg/g.And the highest content of adenosine which was 5.24 mg/g was found in fruiting body of variety B21 inoculated with strain H71.Contents of cordycepin,pentostatin and adenosine in stiff larva were extremely significant lower(P<0.01)than those in fruiting body from all testing groups.The highest contents of cordycepin,pentostatin and adenosine in stiff larva were found in variety B21 inoculated with strain H22,which were 0.39 mg/g,2.67 mg/g and 1.10 mg/g,respectively.Therefore,it can be preliminarily concluded that Philosamia cynthia ricini variety B2 inoculated with Cordyceps militaris strain H22 has a promising future in the production of pentastatin and cordycepin for medical use.
