摘要 蚕丝蛋白具有诸多优良特性,在美容护肤、食品、化工、医药等领域应用广泛。由于丝胶蛋白和丝素蛋白的分子结构不同,在使用前常采用不同的方法将它们分离,存在回收困难、能耗高等问题。研究影响NaOH-尿素法快速溶解蚕丝(丝素和丝胶)的关键因素,获得最佳溶解条件为:在1 L溶解体系中,40 g蚕茧在NaOH浓度为1.20 mol/L、尿素浓度为8 mol/L、温度为40℃的条件下水解3 h可完全溶解。所制备蚕丝蛋白的酪氨酸酶抑制率、亚铁离子螯合能力极显著高于单一的丝胶蛋白和丝素蛋白,其保湿性能与丝素蛋白基本一致。用NaOH-尿素体系溶解蚕丝蛋白,操作简便,反应环境温和,产物具有良好的美白和抗氧化特性。 Silk protein with various advantages is widely used in the fields of cosmetics,food,chemical industry,and medical industry,etc.Due to different molecular structure between sericin and fibroin,varied separation methods in silk dissolving process result in problems like low separation rate and high energy consumption.In this paper,key factors were studied in the process of dissolving silk protein(separating sericin from fibroin)with NaOH-Urea method to acquire the optimized conditions:40 g silkworm cocoons could be completely dissolved in the 1 L dissolving system with NaOH concentration of 1.20 mol/L and urea concentration of 8 mol/L at a temperature of 40℃ for 3 h.Prepared silk protein showed significantly higher inhibition rate of tyrosinase and stronger chelation ability to ferrous ion than those of either fibroin or sericin,and had similar moisture retention ability to fibroin.Dissolving silk with NaOH-Urea method is superior in easy processing under moderate reaction condition,and the product has the skin care characteristics of whitening and anti-oxidation.
