火针对盐酸氨酮戊酸体外经皮渗透的比较

摘要 目的研究火针对促进盐酸氨酮戊酸透皮吸收的影响。方法将24只3~5 d的SD幼鼠随机分为4组,分别为:空白对照组,火针组,普通针组,CO2点阵激光组(n=6)。通过病理组织切片和染料结合试验观察各组在SD幼鼠皮肤中产生的微孔道,检测不同时间点SD幼鼠皮肤的经皮水分丢失率分析皮肤屏障受损情况,再通过经典的垂直透皮扩散试验结合高效液相色谱法计算盐酸氨酮戊酸的体外透皮累积量。结果 HE染色可见火针穿刺深,达(1 215±91.65)μm,孔道闭合快。经皮水分丢失率显示火针组相对CO2点阵激光组,受损皮肤屏障修复更快(P<0.001);累积透皮量结果显示,火针对盐酸氨酮戊酸的促渗作用相比普通针组及CO2点阵激光组更具有明显优势(P<0.001)。结论盐酸氨酮戊酸经火针透皮给药后,渗透率显著提高,且皮肤屏障损伤小,修复快。 Objective To investigate the effect of a fire needle on promoting transdermal absorption of aminolevulinic acid(ALA).Methods Twenty-four SD rats aged 3~5 days were randomly divided into four groups:control group(no intervention),fire needle group,normal needle group,CO2 fractional laser group(n=6).The micropores produced in the skin of SD rats were observed by pathological tissue section and dye-binding experiment.The damage of the skin barrier was measured by detecting the transepidermal water loss at different time points.The classical vertical transdermal diffusion experiment combined with high-performance liquid chromatography was used to calculate in vitro transdermal accumulation of ALA.Results HE staining showed that the puncture depth of fire needles was up to(1215±91.65)μm,and the tunnels closed quickly.Transepidermal water loss data showed that skin in fire needle group repaired faster than in the CO2 fractional laser group(P<0.001).The results of the cumulative transdermal volume showed that the penetration-promoting effect of the fire needle had obvious advantages compared with normal needle group and CO2 fractional laser group(P<0.001).Conclusion After transdermal administration by fire needle,the ALA permeability is significantly improved,the skin barrier is less damaged and repairs quickly.
