宣肺平喘方干预对哮喘大鼠肺、肠组织病理变化及肠道微生物的影响 被引量:6

Effects of Xuanfei Pingchuan Decoction(宣肺平喘方)on Pathological Changes of Lung and Intestine Tissue and Enteric Microorganisms in Asthmatic Rats

摘要 目的:研究宣肺平喘方干预对哮喘大鼠肺、肠组织病理变化及肠道菌群变化的影响,初步探讨哮喘肠道微生物的组成,为后续从微生物角度干预哮喘打下基础。方法:将40只SD大鼠,随机分为正常组、模型组、地塞米松组、宣肺平喘方组,每组10只。通过腹腔注射OVA与氢氧化铝致敏、OVA雾化吸入激发制备哮喘大鼠模型。采用HE染色评价肺、肠组织病理变化,16sRNA测序分析各组大鼠肠道微生物的变化。结果:正常组肺、肠组织均未见炎性病理改变;模型组肺部可见典型支气管哮喘病理表现,肠组织可见炎细胞浸润;用药治疗后炎症等病理变化缓解明显。16sRNA测序分析显示各组大鼠肠道均含有大量的微生物,以模型组最多,正常组及药物组减少。结论:宣肺平喘方能减轻哮喘大鼠肺、肠组织病理损伤;且调节肠道致病菌群,提示其可能从微生物角度抑制哮喘的发作。 Objective:To study the effects of Xuanfei Pingchuan Decoction(宣肺平喘方)on pathological changes of lung and intestine tissue and enteric microorganisms in asthmatic rats,and explore the composition of intestinal microorganisms in asthma,so as to lay a foundation for the follow-up intervention of asthma from the perspective of microorganisms.Methods:40 SD rats were randomly divided into normal group,model group,Dexamethasone group and Xuanfei Pingchuan Decoction group,each with 10 rats.Asthma rat models were induced by intraperitoneal injection of OVA,sensitization with aluminium hydroxide and inhalation of OVA.HE staining was used to evaluate the pathological changes of lung and intestine tissue,and 16sRNA sequencing was used to analyze the changes of intestinal microorganisms in each group.Results:No inflammatory pathological changes were found in the lung and intestine tissues of the normal group;typical bronchial asthma pathological manifestations were observed in the lungs of the model group,and inflammatory cells infiltrated in the intestinal tissues;the pathological changes such as inflammation alleviated significantly after treatment with drugs;16sRNA sequencing analysis showed that there were a large number of microorganisms in the intestine of rats in each group,most in the model group,and less in the normal group and medicine groups.Conclusion:Xuanfei Pingchuan Decoction can alleviate the pathological damage of lung and intestine tissue in asthmatic rats,and it can regulate the pathogenic bacteria of gastrointestinal tract,infer that it may inhibit the onset of asthma from the microbial point of view,and create an early basis for the later use of microorganisms to treat asthma.
