Establishment of Liver Metastasis Model of Gastric Cancer by Spleen Injection of BGC-823 Cells

目的:建立一种符合临床特征的、稳定的胃癌肝转移模型。方法:选用人胃癌细胞BGC-823,转染荧光素酶后通过脾脏注射入CB-17 SCID雄性小鼠,然后切除脾脏。分别在7、14、30和60 d观察小鼠状态以及使用小动物活体成像仪观察肿瘤大小,随后在第30天和第60天分别处死3只小鼠,取肝、胃、肾、肺,HE染色镜下观察这4种脏器的病理学变化。结果:6只小鼠肝脏全部成瘤,成瘤率达到100%,小鼠逐渐出现行动迟缓、摄食量减少、消瘦、精神差、腹部隆起等表现。HE染色可见肝脏转移灶出现坏死。结论:本研究成功建立了人胃癌细胞的肝转移小鼠模型,为后续研究肿瘤行为及抗肿瘤药物奠定基础。

Objective To establish a stable liver metastasis model of gastric cancer that meets clinical characteristics. Methods Human gastric cancer cell line BGC-823 was used, transfected with luciferase and injected into CB-17 SCID male mice through the spleen, and then the spleen was removed. The status of the mice were observed at 7, 14, 30, and 60 days, and the tumor size was observed using a small animal in vivo imager. Then, 3 mice were sacrificed on the 30 th and 60 th days, and the liver, stomach, kidney, and lung were taken. The pathological changes of the four organs were observed under HE staining. Results All the livers of 6 mice were tumorigenic, and the tumor formation rate reached 100%. The mice gradually showed signs of slow action, reduced food intake, weight loss, poor mentality and abdominal bulge. HE staining showed necrosis in liver metastases. Conclusion This study successfully established a mouse model of liver metastasis of human gastric cancer cells, which lays a foundation for subsequent research on tumor behavior and anti-tumor drugs.
