摘要
目的探讨人卵巢颗粒细胞随着年龄增长是否明显衰老。方法在患者取卵过程中收集颗粒细胞,利用Real-time PCR方法检测其衰老相关基因Sirt1表达、利用T/S Ratio方法检测端粒长度及体外培养颗粒细胞检测其增殖能力。结果颗粒细胞随着年龄增长衰老相关基因Sirt1表达无明显增长,端粒长度无明显缩短,到45岁颗粒细胞依然保持较强的增殖能力。结论卵子随着年龄迅速衰老,而颗粒细胞未见明显的衰老。
Objective To investigate whether the human ovarian granulosa cells aging obviously as age increasing.Methods Granulosa cells was collected during the process of picking up oocytes.Real-time PCR method was used to detect the expression of Sirt1,and T/S Ratio method to detect the length of telomere and the proliferation ability of granulosa cells in vitro.Results The expression of Sirt1 of ovarian granulosa cells was not significantly increased,and the telomere length did not significantly shorten,as age increasing.The proliferation ability of granulosa cells maintained high till the age of 45 years.Conclusion The ovarian granulosa cells were not aging significantly,although the eggs was aging obviously as age increasing.
