新疆沙雅县图兰扇头蜱的形态学及分子生物学鉴定 被引量:1

摘要 为了掌握新疆边境地区沙雅县的蜱种分布状况,从该地绵羊体表采集寄生蜱虫,应用数码体视显微镜对其进行形态学初步鉴定,采用分子生物学技术对采集蜱虫的线粒体16S rDNA进行基因序列扩增并测序,应用序列分析软件MEGA 6.0构建系统发育进化树,分析采集蜱样的分子进化特征。结果表明,采集的蜱样均为图兰扇头蜱,在系统发育上依然和新疆境内石河子地区、乌什地区、皮山地区和图木舒克地区的图兰扇头蜱处于同一分支,具有分子进化的相对保守性。 In order to grasp the distribution of tick species in Shaya county in the border area of Xinjiang,parasitic ticks were collected from the surface of the sheep in the area,and their morphology was preliminarily identified by digital stereo microscope.The mitochondrial 16S rDNA of ticks was amplified and sequenced by molecular biology method.The phylogenetic tree was constructed by using sequence analysis software MEGA 6.0 to analyze the molecular evolution characteristics of the collected samples.The results showed that the samples collected in this study were all Rhipicephalus turanicus,and they were still in the same branch of Rhipicephalus turanicus in Shihezi,Wushi,Pishan and Tumushu areas in Xinjiang,with relative conservation of molecular evolution.
