摘要
[目的]为了研究有机磷化合物在体外对猪肾细胞损伤作用。[方法]选择敌敌畏和马拉硫磷作用猪肾细胞系PK15。MTT比色法测量2种有机磷化合物对细胞抑制作用,荧光染色和DNA ladder法检测它们对PK15的致凋亡作用。同时检测氧化相关因子超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量和PON1基因m RNA水平。[结果]敌敌畏和马拉硫磷抑制PK15细胞的生长呈时间-剂量效应,并能诱导PK15产生较明显的凋亡现象,同时能使细胞中的超氧化物歧化酶活性下降、丙二醛水平上升。[结论]2种有机磷农药可使细胞的抗氧化能力下降,致使细胞中的氧自由基水平上升,可能是有机磷化合物致细胞凋亡、损伤的因素之一。
[Aims]To explore the effects of organophosphorus pesticide on the injury of porcine kidney cell.[Methods]PK15 cells were treated with dichlorvos and malathion,respectively.The proliferation of PK15 cells was analyzed by MTT assay and the apoptosis was detected by acridine orange-ethidium bromide(AO/EB)double fluorescent dye staining and DNA ladder methods.Such oxidation stress related factors:superoxide dismutase(SOD),malondialdehyde(MDA)and PON1 mRNA levels were also measured.[Results]The results showed that both of dichlorvos and malathion could induce PK15 apoptosis.The activity of SOD and PON1 mR NA were reduced and MDA was increased when the PK15 cells were exposure under the two kind of pesticides.The results implied the reactive oxygen species(ROS)of cellular was raised by dichlorvos and malathion.[Conclusions]It suggested that the two organophosphorus pesticides can reduce cellular antioxidant ability and raise the level of ROS,which is one of the reasons to induce the injury and apoptosis of swine kidney cells.
