摘要
以Sinularia软珊瑚为研究对象,用Covaris超声波破碎仪随机将其DNA样品打断成长度为350 bp的片段,构建好的文库通过Illumina Nova Seq进行PE测序.结果表明:软珊瑚Sinularia ceramensis Verseveldt基因组大小为649. 58 Mbp,杂合率为1. 25%,重复序列比例为57. 06%.采用kmer=43进行初步组装,得到contig N50为836 bp,总长为586. 07 Mbp,scaffold N50为1 044 bp,总长为599. 11 Mbp.结果显示,Sinularia ceramensis Verseveldt的基因组为1个复杂的基因组,需要采用相应的策略进行基因组组装.
In this research the qualified DNA of Sinularia soft coral-the subject of research-was randomly interrupted with 350 bp fragments by Covaris ultrasonic breaker to construct a library which was sequenced by PE by using Illumina Nova Seq. The genome size of Sinularia ceramensis Verseveldt was 649. 58 Mbp,of which the heterozygosity rate was 1. 25%,and the repeat sequence proportion was 57. 06%. Preliminary assembled with kmer = 43,the contig N50 was 836 bp with a total length of 586. 07 Mbp and the scaffold N50 was 1,044 bp with a total length of 599. 11 Mbp. The result showed that the sequenced genome is a complex one,therefore the corresponding strategies are needed to be used for genome assembly.
