产细菌素肠球菌的分离及其抑菌谱的筛选

摘要 目的:本试验从大鼠等7种健康动物粪便中分离出7株肠球菌,提取细菌素并进行抑菌谱的分析,为细菌素的研究提供参考。方法:采集大鼠、小鼠、黄牛、奶牛、山羊、家兔、鸡等7种健康动物的粪便,用肠球菌选择培养基进行肠球菌的分离培养,对分离到的典型菌落进行革兰染色镜检,分别用LB液体培养基对7株分离菌各培养48 h后,制备发酵上清液,然后用硫酸铵盐析法制备粗提细菌素,采用圆纸片扩散法对27株细菌抑菌试验,筛选抑菌谱。结果:从上述7种健康动物的粪便中分离出7株肠球菌,肠球菌的发酵上清液对不同株肠球菌、大肠杆菌、白色葡萄球菌等细菌有抑制作用,肠球菌细菌素不仅对白色葡萄球菌等革兰阳性菌具有抑制作用,同时对大肠杆菌、沙门氏菌、产气杆菌等革兰阴性菌也表现出抑制作用。结论:肠球菌的发酵上清液及细菌素均有抑菌作用,具有较广的抑菌谱。 Objective:The test from the feces of seven kinds of animal health isolated 7 strains of enterococci were screened and bacteriocin extraction and screening of antibacterial spectrum,for bacteriocin research provide reference.Methods:Acquisition in rats,mice,cattle,dairy cattle,goats,rabbits 7 healthy animal feces,with Enterococcus selective medium of enterococci isolated culture was carried out.The isolated typical colonies of gram staining microscopic examination,with LB liquid medium for bacteriocin production.After cultured for 48 h,preparation of fermentation supernatant,and with ammonium sulfate salting out method of bacteriocin crude extract respectively,and on 26 strains of bacteria with circular slip method were tested for antimicrobial activity,screening antibacterial spectrum.Results:7 strains of enterococci isolated from the seven kinds of animal health of feces and Enterococcus faecium fermentation supernatant of different strains of Enterococcus,Escherichia coli,Staphylococcus aureus bacteria have inhibitory effect,enterococcal bacteriocin not only on Staphylococcus aureus and other leather of gram positive bacteria have inhibitory effect,also on Escherichia coli,Salmonella,gas producing Bacillus leather gram negative bacteria also showed inhibito.Conclusion:In this experiment,the fermentation supernatant of Enterococcus and bacteriocins have antimicrobial effect,has a wider antibacterial spectrum.
