摘要
利用超高效液相串联质谱法(UPLC-MS/MS)对牛肉中牛血清白蛋白(BSA)和马肉中马血清白蛋白(ESA)经胰蛋白酶消化后的目标肽进行鉴定,采用多反应监测(MRM)方法对结果进行验证,建立了一种检测牛肉中掺杂马肉的方法.对UPLC-MS/MS数据的深入研究表明,共检测到9个牛特异性多肽和17个马特异性多肽.其中DAFLGSFLYEYSR(m/z 784.3,Charge 2+)和ADFAEVSK(m/z 433.7,Charge 2+)的质谱响应最好,被选为牛肉和马肉定量的靶肽.所建方法在5.5~550.0和4.875~487.5 mg/L范围内线性关系较好(r^2>0.99),检测限、选择性、精密度、回收率和稳定性完全满足分析要求.
An optimized method was developed for the detection of horse meat in beef. Target peptides from tryptic-digested bovine serum albumin(BSA) and equine serum albumin(ESA) of beef and horse meat were identified using ultra-performance liquid tandem mass spectrometry(UPLC-MS/MS). A multiple reaction monitoring(MRM) method was established for verification. A thorough investigation of the UPLC-MS/MS data showed that 9 bovine-specific and 17 equine-specific peptides were detected. Among them, the peptides DAFLGSFLYEYSR(m/z 784.3, charge 2+) and ADFAEVSK(m/z 433.7, charge 2+) had the best MS responses and were chosen as the target peptides for quantification of beef and horse meat. The method was linear or polynomial(r^2>0.99) for BSA and ESA in the range of 5.5-550.0 and 4.875-487.5 mg/L. The detection limit, selectivity, precision, recovery and stability could completely meet the analytic requirements.
