摘要
目的探讨胃癌组织中长链非编码RNA(LncRNA)HLA复合体11(HCG11)及微小RNA(miR)-144-3p的表达及临床意义。方法应用qRT-PCR检测102例胃癌组织和癌旁正常胃组织中LncRNA HCG11及miR-144-3p的表达,比较两种组织间LncRNA HCG11、miR-144-3p表达差异,分析胃癌组织LncRNA HCG11、miR-144-3p表达与患者临床病理特征及预后的关系。结果与癌旁正常组织相比,胃癌组织中LncRNA HCG11表达升高,而miR-144-3p降低(P均<0.05)。癌组织中LncRNA HCG11、miR-144-3p表达水平与肿瘤TNM分期及组织学分化有关(P均<0.05),与性别、年龄、肿瘤直径、位置及淋巴结转移情况无关(P均>0.05)。癌组织中LncRNA HCG11与miR-144-3p呈负相关(r=-0.611,P<0.05)。高LncRNA HCG11表达组患者3年总体生存率(OS)低于低LncRNA HCG11表达组患者,低miR-144-3p表达组患者3年OS低于高miR-144-3p表达患者(P均<0.05)。结论胃癌组织中LncRNA HCG11表达升高、miR-144-3p表达降低,二者均与胃癌发生发展相关,有望成为新的胃癌肿瘤标志物。
Objective To study the expression and clinical significance of long non-coding RNA(LncRNA)HLA complex group 11(HCG11)and microRNA(miR)-144-3p in the gastric cancer tissues.Methods qRT-PCR was used to detect the expression levels of LncRNA HCG11 and miR-144-3p in 102 gastric cancer tissues and adjacent normal gastric tissues.The expression differences in LncRNA HCG11 and miR-144-3p between the two tissues were compared.The relationships between the expression of LncRNA HCG11 and miR-144-3p and the clinicopathological characteristics and prognosis of patients were analyzed.Results Compared with the adjacent normal tissues,the expression of LncRNA HCG11 in the gastric cancer tissues increased,while miR-144-3p decreased(all P<0.05).The expression levels of LncRNA HCG11 and miR-144-3p in the cancer tissues were related to tumor TNM stage and histological differentiation(both P<0.05),but not to gender,age,tumor diameter,location or lymph node metastasis(all P>0.05).LncRNA HCG11 was significantly negatively correlated with miR-144-3p in the cancer tissues(r=-0.611,P<0.05).The 3-year overall survival(OS)of patients with high LncRNA HCG11 expression was significantly lower than that of patients with low LncRNA HCG11 expression,and the 3-year OS of patients with low miR-144-3p expression was lower than that of patients with high miR-144-3p expression(both P<0.05).Conclusion The expression of LncRNA HCG11 in gastric cancer tissues increases,while the expression of miR-144-3p decreases;both of them are involved in the occurrence and development of gastric cancer,and are expected to become new tumor markers for gastric cancer.
