摘要
目的探讨lncRNA MALAT1在糖尿病新生血管性青光眼虹膜组织中的表达及意义。方法糖尿病新生血管性青光眼患者92例为病例组,同期虹膜无新生血管的慢性闭角型青光眼患者92例为对照组。2组在行小梁切除术行虹膜根切时留取虹膜组织,测定虹膜组织lncRNA MALAT1 mRNA、蛋白及白介素-2(IL-2)、白介素-6(IL-6)、超敏-C反应蛋白(hs-CRP)、肿瘤坏死因子-α(TNF-α)水平。结果病例组lncRNA MALAT1蛋白表达评分、阴性表达率、MRNA表达水平高于对照组(P<0.01);病例组lncRNA MALAT1蛋白表达阳性率明显高于对照组(P<0.01);病例组IL-2、IL-6、hs-CRP、TNF-α水平高于对照组,差异有统计学意义(P<0.01);lncRNA MALAT1蛋白表达评分、lncRNA MALAT1 mRNA与IL-2、IL-6、hs-CRP、TNF-α呈正相关(r值分别为0.651、0.572、0.483、0.494、0.636、0.718、0.509、0.520,P<0.05)。结论lncRNA MALAT1在糖尿病新生血管性青光眼虹膜组织中表达水平升高,其可诱导炎症反应加剧糖尿病新生血管性青光眼的病变程度。
Objective To investigate the expression and significance of lncRNA MALAT1 in iris tissues of patients with diabetic neovascular glaucoma.Methods A total of 92 patients with diabetic neovascular glaucoma were enrolled as case group,and the other 92 cases patients with chronic angle-closure glaucoma without iris neovascularization were enrolled as control group.The iris tissues were collected during trabeculectomy,and the levels of lncRNA MALAT1 mRNA and protein,IL-2,IL-6,hs-CRP,TNF-αwere detected and compared between the two groups.Results The protein expression levels of lncRNA MALAT1,the positive rate of MALAT1 protein expression and mRNA expression levels of MALAT1 in case group were significantly higher than those in control group(P<0.01).The levels of IL-2,IL-6,hs-CRP and TNF-αin case group were significantly higher than those in control group(P<0.01)。Moreover the mRNA and protein expression scores of lncRNA MALAT1 were positively correlated with the levels of IL-2,IL-6,hs-CRP and TNF-α(P<0.05).Conclusion The expression levels of lncRNA MALAT1 are increased in iris tissues of patients with diabetic neovascular glaucoma.which can induce the inflammatory reaction to aggravate the pathological change severity of diabetic neovascular glaucoma.
